黄芪多糖在宿主抵抗李斯特菌中的作用

摘    要:目的:观察黄芪提取物注射小鼠后,对小鼠抵抗单核细胞增生李斯特菌能力的影响。方法:水煮法提取黄芪多糖,配成悬液,腹腔注射感染李斯特菌的小鼠,并设对照,每组20只昆明鼠,观察小鼠的生存时间,脾脏的重量指数,脏器菌落计数,检测小鼠血清中抗体IgG水平,利用ELISA法测定小鼠脾脏释放的细胞因子。结果:注射黄芪多糖的小鼠,在30d观察期结束后仍有12只存活,而对照组只有2只存活。黄芪多糖组小鼠脾脏的重量指数(5.2±1.2),菌落计数(4.1±1.5)均比对照组的重量指数(11.0±0.9),菌落计数(8.8±1.3)明显降低。黄芪多糖组小鼠血清中抗体滴度(OD490)为0.38±0.05,对照组的抗体滴度为0.18±0.07。ELISA法测的黄芪多糖组小鼠脾脏细胞分泌IFN-γ为(240±15)ng/L,对照组为(150±19)ng/L。结论:黄芪多糖能够增强宿主的体液免疫和细胞免疫来保护宿主抵抗胞内菌的感染。

Abstract:Objective: To demonstrate whether astragalus mongholicus polysaccharide(APS) would suppress L.monocytohenes infection in mice. Methods: APS was extracted with boiling methods. Kunming mice were randomized into PBS group (control group) and APS group equally. APS group were treated with APS via intraperitoneally injection for 3 days successively, then every mouse was infected with L.monocytohenes. The survival rate of mice were recorded, numbers of viable bacteria in the spleen were counted and the weight index of spleen (WI),levels of IgG in mouse sera and levels of IFN-γ in mouse spleens were measured. Results: In APS group,survival rate (60%)of mice was higher than in PBS group(10%), and the numbers of viable bacteria (4.1±1.5) in spleen and WI(5.2±1.2) of spleen were also much lower than in PBS group(8.8±1.3 and 11.0±0.9, respectively). The IgG titer(OD490) was (0.38±0.05) and IFN-γ level was(240±15)ng/L, and was much higher than in PBS group, as (0.18±0.07) and (150±19)ng/L, respectively. Conclusion: Astragalus mongholicus polysaccharide can protect host against L.monocytohenes infection by enhancing humoral immunity and cellular immunity.
