摘    要:目的:探讨黄芪多糖(APS)对2型糖尿病小鼠(KKAy)早期肾脏病理改变的影响。方法:雌性KKAy小鼠和C57BL/6J小鼠随机分为:糖尿病组(DM,n=8)、糖尿病黄芪多糖治疗组(DA,n=8)、正常对照组(C,n=10)和黄芪多糖对照组(CA,n=10)。定期监测体重、血糖、血胰岛素水平、计算HOMA-IR指数;观察肾小球病理变化。结果:KKAy鼠较C57BL/6J鼠体重、血糖、血胰岛素及HOMA-IR均显著升高(P<0.01)。DM组小鼠肾小球面积较C组增大,系膜基质增加、肾小管管腔扩大、管壁变薄、肾小球基底膜(GBM)增厚、蛋白管型明显。经APS治疗后,以上指标均明显改善(P<0.01)。结论:APS可降低血糖,增强机体对胰岛素敏感性,对2型糖尿病小鼠早期肾脏病理改变具有良好的治疗作用。

Abstract:Objective: To investigate the effects of Astragalus Polysaccharide(APS) on renal pathological changes in the early period of type 2 diabetic KKAy mice model.Methods: Female KKAy and C57BL/6J mice were assigned into four groups randomly: diabetes(DM) group(n=8);diabetes with APS therapy(DA) group(n=8);control(C) group(n=10) and APS control(CA) group (n=10).Body weight,blood glucose was detected.Insulin sensitivity was analyzed by blood insulin level and HOMA IR index was calculated.Early renal pathological variation was presented by histochemical staining and TEM.Results: After APS therapy for 8 weeks,the level of blood glucose,blood insulin,body weight and HOMA IR index was decreased in DA group(P<0.01,vs DM group).In DM group mice,renal glomerulus area was magnified,mesenterium basilaris substantia was increased,urinary cast lumina was enlarged,its vessel wall was thinner,GBM was thickening and obvious protein casts were observed compared with those of C group.APS treatment can alleviate these pathological changes.Conclusion: The results indicate that APS can ameliorate the renal pathological variation in the early period,by its effects of insulin-sensitizing and hypoglycemic activity.
