SPA结合HSV免疫血清交叉吸收抗原后对抗体分型的初步探讨

以HSV-1(F株)或HSV-2(G株)免疫血清结合SPA,然后用于交叉吸收HSV-2或HSV-1抗原,经吸收的抗原作ELISA,目的是检测HSV型特异性抗体。在未经吸收的抗原反应中,八份兔抗血清的同型反应与交叉反应差别不大,其抗体效价比值在2倍以内,而经交叉吸收的抗原反应中,抗体以同型反应为主,其抗体效价比值都在4倍以上,最高达16倍。经多次交叉吸收抗原后,抗体的交叉反应进一步降低,但其同型反应也有所降低。

Study of HSV Type-Specific Antibodies by Cross-Absorbation with SPA Coasted with Anti-HSV Rabbit Immunoseia

The HSV-1 antigen (or HSV-2 antigen) was across absorbed by SPA coasted with anti-HSV-2 (or anti-HSV-1) rabbit immunoserum. The absorbed antigens were used in ELISA for detecting the type-specific antibodies to HSV-1 or HSV-2. When reared with the unabsorbed antigens, the eigth rabbit immunosera did not show the difference between the homoreaclion and the cross-reaction. But when reated with the absorbed antigens, these immunosera mainly showed the homoreaction. The ratios of the titers of the homoreactive antibodies in these sera to those of the cross-rective antibodies were more than 4 times, the highest reaching the 16 times. When the reaptedly absorbed antigens were used in ELISA. the strength of cross-reactive antibodies was greatly lowered, but homoreaction was some what weakened also. The discussions were made about the experimental method and the type-specific antibody reactions to HSV antigens.
