摘    要:目的 探讨长链非编码RNA MEG3(lncRNA MEG3)在2型糖尿病(T2DM)伴幽门螺杆菌(Hp)感染患者血清中的表达及意义。方法选取132例T2DM患者为研究对象,根据是否感染Hp分为非感染Hp组(49例)和感染Hp组(83例)。实时荧光定量PCR(RT-qPCR)检测两组患者血清lncRNA MEG3水平,全自动分析仪检测血清游离脂肪酸(FFA)水平,乳胶增强免疫(超敏)比浊法检测超敏C反应蛋白(hs-CRP)、酶联免疫吸附法检测肿瘤坏死因子α(TNF-α)、白细胞介素-6(IL-6)、丙二醛(MDA)水平、超氧化物歧化酶(SOD)和谷胱甘肽还原酶(GSH-Px)表达水平。比较两组患者各检测指标水平差异,并进行Pearson相关性分析。Logistic回归分析影响T2DM患者感染Hp的因素。结果与非感染Hp组比较,感染Hp组患者血清lncRNA MEG3、SOD和GSH-Px水平显著降低(P<0.05),FFA、TNF-α、IL-6、hs-CRP和MDA水平均显著升高(P<0.05)。Pearson相关性分析结果显示,T2DM伴Hp感染患者血清lncRNA MEG3水平与FFA、TNF-α、IL-6、hs-CRP和MDA呈负相关(P<0.05),与SOD和 GSH-Px呈正相关(P<0.05)。Logistic多因素分析结果显示,lncRNA MEG3是影响T2DM患者感染Hp的独立因素(P<0.05)。结论lncRNA MEG3在T2DM伴Hp感染患者血清中表达降低,其可能通过调节机体内脂肪酸水平、炎性因子表达及氧化应激水平参与T2DM感染Hp过程。

Abstract:Objective To investigate the expression and significance of lncRNA MEG3serum of T2DM patients infected with Hp. Methods 132 T2DM patients were selected. They were divided into non-Hp group (49 cases) and Hp group (83 cases) according to whether they were infected with Hp. RT-qPCR was used to detect the serum lncRNA MEG3 levels. The fully automatic analyzer was used to detect serum free fatty acid (FFA) levels. The latex enhanced immune (hypersensitivity) turbidimetric method was used to detect the hs-CRP. Enzyme-linked immunosorbent assay was used to detect the levels of TNF-α, IL-6, MDA, SOD and GSH-Px. The differences in the levels of detection indicators between the two groups of patients were compared, and Pearson correlation analysis was performed. Logistic regression analysis was performed to analyzed the factors affecting Hp infection in T2DM patients. Results Compared with the non-Hp group, serum levels of lncRNA MEG3, SOD and GSH-Px in patients with Hp infection were significantly reduced (P<0.05), and levels of FFA, TNF-α, IL-6, hs-CRP and MDA were significantly increased (P<0.05). Pearson correlation analysis showed that serum lncRNA MEG3 level in T2DM patients infected with Hp was significantly negatively correlated with FFA, TNF-α, IL-6, hs-CRP, and MDA (P<0.05), and positively correlated with SOD and GSH-Px (P<0.05). Logistic multivariate analysis showed that lncRNA MEG3 and hs-CRP were independent factors affecting Hp infection in T2DM patients. Conclusion The expression of lncRNA MEG3 is reduced in serum of T2DM patients infected with Hp. It may participate in the process of Hp infection in T2DM by regulating the level of fatty acids, inflammatory factors and oxidative stress in the body.
