当归诱导骨髓间充质干细胞分化为神经元样细胞

摘    要:目的:研究当归体外定向诱导大鼠骨髓间充质干细胞(MSCs)分化的作用。方法:通过贴壁法分离大鼠MSCs,体外扩增培养。MSC用20μl当归注射液/ml含10%胎牛血清的L—DMEM为预诱导条件,预诱导24h后用当归注射液100μl/ml不含胎牛血清的L—DMEM进行诱导。光镜下观察细胞形态,免疫细胞化学检测神经细胞特异性抗原标志。结果:大鼠骨髓间充质干细胞可通过贴壁法成功分离并可在体外大量扩增。当归诱导3h后大部分MSCs转变为神经元样细胞,出现胞体和突起,免疫细胞化学染色NSE及nestin呈阳性、GFAP性。结论:大鼠骨髓间质干细胞可在体外诱导分化为神经元样细胞。

Abstract:Objective To investigate the differentiation from rat mesenchymal stem cells(MSCs) to neuron-like cells with Chinese angelica. Methods Mesenchymal stem cells were obtained from thighbones marrow of rats and expended in culture medium. Chinese angelica was used to study its differentiative effect on MSCs. MSCs were pretreated with preinduction medium consisting of L-DMEM/10%FCS/2%(V/V) Chinese angelica for 24 hours, then were induced with induction media consisting of L-DMEM/2%(V/V) Chinese angelica. Neuron-specific enolase(NSE), nestin, gliall fibrillary acaidic protein(GFAP) were detected by immunocytochemistry. Results Rat MSCs were expanded as undifferentiated cells in culture for more than 20 passages. MSCs were induced with simple Chinese angelica, which could generate neuron like-cells within 3 hours. The expression of NSE and nestin in the neuron-like cells was positive but GFAP was negative. Conclusion MSCs can be induced to differentiate into neuron like-cells with Chinese angelica.
