胃癌组织中COX-2与P300/CBP表达的研究

摘    要:目的:研究胃癌组织中环氧化酶-2(COX-2)和核蛋白P300/CBP的表达变化,并探讨两者与胃癌生物学行为的关系。方法:用免疫组织化学S-P法检测不同分化程度胃癌组织和正常胃黏膜组织中COX-2和P300/CBP的表达变化。结果:(1)COX-2在胃癌组织的表达阳性率为37.8%,明显高于正常胃黏膜组织(0.0),P<0.05;在低分化型胃癌阳性率为56.1%,明显高于高中分化型胃癌组织(8.0%),P<0.05。(2)P300/CBP在胃癌组织的表达阳性率为10.6%,明显低于正常胃黏膜组织(72.2%),P<0.05;在低分化型胃癌阳性率为4.9%,低于高中分化型胃癌组织(20.0%),但P>0.05,差异无显著性。(3)COX-2表达阳性率在胃癌淋巴结转移组和远处转移组均显著高于非转移组,P<0.05;P300/CBP表达阳性率在胃癌的淋巴结转移组和远处转移组均低于非转移组,但P>0.05,差异无显著性。(4)胃癌组织中COX-2和P300/CBP表达呈现负相关倾向。结论:COX-2和P300/CBP在胃癌组织中表达异常,在胃癌的发生、发展中起重要作用,可作为辅助临床判断胃癌分化程度和转移生物学行为的有用指标。

Study on expression of COX-2 and P300/CBP in gastric carcinoma
Abstract:Aim: To study the expression of COX-2 and P300/CBP in gastric carcinoma,and to investigate their relationship with the biological behaviors of gastric carcinoma. METHODS: The expression of COX-2 and P300/CBP in normal gastric mucosa and gastric carcinoma was examined by S-P immunohistochemical staining. RESULTS: (1)The expression rate of COX-2 in gastric carcinoma was 37.8%,which was significantly higher than that in normal gastric mucosa(0%),P<0.05. The expression rate of COX-2 in poorly differentiated gastric carcinoma was 56.1%,which was significantly higher than that in well-moderated differentiated gastric carcinoma (8.0%), P<0.05.(2) The expression rate of P300/CBP in gastric carcinoma was 10.6%,which was significantly lower than that in normal gastric mucosa(72.2%),P<0.05. The expression rate of P300/CBP in poorly differentiated gastric carcinoma was 4.9%,which was lower than that in well-moderated differentiated gastric carcinoma (20.0%) ,but not significantly with P>0.05.(3) There was significant difference in the expression of COX-2(P<0.05) between the group with lymphnode metastasis(76.9%) and the group without lymphnode metastasis(7.4%),and between the group with distant metastasis(76.9%) and the group without distant metastasis(13.2%); There was no significant difference in the expression of P300/CBP(P>0.05) between the group with lymphnode metastasis(5.1%) and the group without lymphnode metastasis(18.5%),and between the group with distant metastasis(7.7%) and the group without distant metastasis(11.3%);(4) There was negative correlative tendency between COX-2 and P300/CBP in gastric carcinoma. CONCLUSION: COX-2 and P300/CBP play important roles in the genesis and development of gastric carcinoma, and can be used as important indexes in the research of the biological behavior and prognosis of gastric cancer.
