系统性红斑狼疮患者外周血中差异表达微小RNAs的筛选

目的通过比较微小RNAs(microRNAs,miRNA)在系统性红斑狼疮(SLE)患者和正常健康对照者外周血单个核细胞(PBMC)中的表达情况,发现在SLE中异常表达的miRNA。方法提取23例SLE患者及10例正常健康对照者PBMC中总RNA并分离miRNA,采用含1 152条探针的哺乳动物miRNA芯片对两者之间差异表达的miRNA进行分析。随机选择其中4个miRNA,应用Northern blot对芯片结果进行验证。结果 SLE患者与正常健康对照者PBMC间差异表达的miRNA共14个,其中8个表达上调,6个表达下调。Northern blot验证结果与芯片表达结果一致。结论多种miRNA在SLE患者PBMC中异常表达,提示它们可能在SLE的发生发展中起着重要的调控作用。

Identification of differentially expressed microRNAs in peripheral blood mononuclear cells of systemic lupus erythematosus patients

Objective To investigate the differentially expressed microRNAs(miRNA) in peripheral mononuclear cells(PBMCs) in the systemic lupus erythematosus(SLE) patients.Methods PBMCs were isolated from blood samples of 23 SLE patients and 10 health controls.The miRNA extracted from PBMC was applied to the miRNA microarray chip.Four random differentially expressed miRNAs were verified by Northern blot analysis.Results There were 14 miRNAs differentially expressed were indentified in SLE according to microarray chip.These results were confirmed by Northern blot analysis.Conclusion The altered expression profile of miRNA in PBMCs of SLE patients implies differentially expressed miRNA plays roles in the pathogenesis of SLE.
