摘    要:目的:用共聚焦定量分析方法,观察拉米夫定脂质纳米粒对细胞内HBV基因表达的作用。方法:构建半乳糖修饰的十六酸拉米夫定脂质纳米粒(LAP-GSLN),将其定向导入2.2.15细胞,对导入的细胞所表达HBeAg进行间接免疫荧光染色,用共聚焦技术对其含量进行定量分析。结果:导入LAP-GSLN的细胞内HBeAg荧光量与单纯加入拉米夫定(LA)比较,导入LAP-GSLN的细胞内HBeAg荧光明明显减少。结论:LAP-GSLN与2.2.15细胞上的半乳糖受体结合,将LA导入细胞而发挥对HBeAg抑制作用。

Abstract:Objective To investigate the effect of LAP-GSLN on HBV expression by laser scanning confocal microscope detection. Methods The nanoparticles modified by galactosides(LAP-GSLN) were prepared and delivered into 2 2 15 cell. Indirect immunofluorescence and laser scanning confocal microscope were used to detect HBeAg expression. Results Compared with cells delivered with LA, the fluorescence intensity in cells delivered with LAP-GSLN was much weaker. Conclusion LAP-GSLN can effectively combine with galactoside receptors and LA can enter cells to inhibit HBeAg expression.
