肺结核患者外周血alpha/betaT细胞CDR3谱系研究

摘    要:摘要] 目的 探讨活动性肺结核患者外周血T细胞TCR α、 链CDR3 谱系漂移,为肺结核患者特异性T细胞免疫应答机制和免疫治疗研究提供基础。方法 采用基因扫描谱型分析技术,分析6例正常献血员及6例活动性肺结核患者PBMC中T细胞TCR CDR3谱型分布特征及患者TCR Vα和Vβ基因家族的优势取用情况,对克隆性增生T 细胞CDR3区进行序列分析。结果 6例正常献血员外周血中TCR 32 Vα、24 Vβ家族CDR3谱型均呈高斯分布(gaussian distribution),6例活动性肺结核患者出现不同Vα和Vβ家族优势取用,对部分单/寡克隆增生T细胞α、 链CDR3区基因进行测序,证实存在不同的CDR3序列。结论 肺结核患者活动期外周血T细胞TCR α、 链CDR3谱系出现明显漂移,提示CDR3的选择性表达与结核患者细胞免疫应答机制有关,优势取用Vα、Vβ家族T细胞TCR CDR3序列的确定,为肺结核患者特异性T细胞免疫治疗研究提供基础。

Analysis of TCR BV CDR3 of αβT cells in the PBMC of patients with active tuberculosis
Abstract:ObjectiveTo analysis the drift of TCR Vαand Vβgene families CDR3 spectratype of T cells in the PBMC of patients with active tuberculosis and provide a basis for the specific T cell immune response mechanism and Immunotherapy research of active tuberculosis. Methods: Immunoscope Spectratyping techniques used to analysis the distribution of six case of normal donation and the advantages usded of TCR αand β chain CDR3 in the six patients with active tuberculosis. Sequence analysis the CDR3 region of monoclonal or oligoclonal T cell. Results: The spectratypes of Vαand Vβ gene familiesˇ TCR CDR3 region of six healthy control donors showed Gaussian distribution. Six patients with active tuberculosis, however, displayed anomalous proliferation and the oligoclonal expansion of T cell were observed at the different TCR Vα and Vβfamilies with different CDR3 sequence. Conclusion: Noticeable drift could discovered for the TCR α and β chain CDR3 in activity period of tuberculosis, which indicate that the advantages usage of TCR Vαand Vβfamilies may be associated with the immune pathogenesis of active tuberculosis. By determining the CDR3 sequence of advantages usage of TCR Vα and Vβ families, we could provide a basis for future research of tuberculosisˇs specific T cell immune therapy.
