Survivin和rac1蛋白在胃癌中的表达及其临床意义

摘要] 目的 探讨Survivin和rac1蛋白在正常胃和胃癌组织中的表达及其与胃癌生物学行为的关系。方法 应用链酶亲和素-生物素过氧化物酶复合物(SABC)免疫组化法检测65例胃癌及癌旁6cm处正常胃黏膜组织中Survivin和rac1蛋白的表达，并分析其与胃癌临床、病理参数之间的关系。结果 (1)Survivin蛋白在癌旁正常胃黏膜组织中染色均为阴性，在胃癌组织中52.3%染色阳性（P<0.01）；rac1蛋白染色阳性率在胃癌组织中(75.4%)显著高于癌旁正常胃黏膜组织（21.5%）（P<0.01）；（2）随胃癌组织分化程度的降低、浸润深度的增加、TNM分期的升高及淋巴结和远处转移的发生, Survivin和rac1蛋白的染色阳性率明显升高(P<0.05)；(3) Survivin阳性胃癌组织中的rac1蛋白表达水平显著高于Survivin阴性者(P<0.01)； 结论 Survivin和rac1的表达与胃癌的生物学行为密切相关，对胃癌组织中Survivin和rac1的联合检测有助于预测胃癌患者的预后。

Expression of surviving and rac1 and clinical significance in gastric carcinoma

Abstract: Objection:To discuss the expression of Survivin and rac1 protein in tissues of normal gastric and gastric carcinoma and its relationship with gastric carcinoma biological benavior.Methods:The expression of survivin and rac1 proteins in 65 cases of gastric carcinoma and paracarcinoma mucosa tissues which were 6 cm from the tumer were detected using Strept Avidin-Biotin Complex (SABC) immunohistochemical method, and the correlation between the expression of survivin,rac1 and the clinical,pathological parame- ters of gastric cancer were analysed .Results: (1)The staining of Survivin protein in normal gastric tissues was negative .While positive staining was detected in gastric carcinoma tissues (52.3%) (p<0.01)；rac1 positive staining rate in gastric carcinoma tissues was 75.4%, while in normal gastric mucosa tissues was 21.5% (p<0.01)；（2）In gastic carcinoma musoca tissues ,As the degree of histologic polarization decreased,depth of invasion increased,stage of TNM was upgraded ,lymph node and metastasis and hematogenous metastasis apprared, the positive staining rate of survivin and rac1 protein was increased (p<0.05)；The positive staining rate of rac1 protein was signify- cantly higher in gastric carcinoma tissues which the Survivin protein is positive than which the Survivin protein is negative (p<0.01)；Conclusions:Positive correlation exsisted between the positive expression of Survivin and rac1 protein and the biological behavior of gastric carcinoma，which suggested that combined examination of Survivin and rac1 in gastric carcinoma can be helpful to forcast its prognosis.
