抗肿瘤中药通光藤的药理作用及其作用机理初步研究

摘    要:目的研究抗肿瘤中药通光藤的药理作用及其作用机理。方法以现有上市制剂消癌平注射液为阳性对照,采用稻瘟霉法和MTT法双重体外筛选模型跟踪筛选其抗肿瘤活性部位,用小鼠Hep 1—6肝癌模型验证活性部位的体内抑瘤药效,进一步采用ELISA法验证通光藤总苷提取物对小鼠腹腔巨噬细胞释放肿瘤坏死因子和小鼠脾细胞生成白细胞介素2的影响。结果双重体外活性筛选结果发现,抗肿瘤中药通光藤的直接细胞毒活性不明显;体内抑瘤药效试验提示,通光藤提取物有明显的抗肿瘤作用,但具有一定的毒性;通光藤的化学部位及其上市对照制剂消癌平注射液对小鼠释放TNF-α和生成IL-2有一定影响,但不同化学部位表现出来的作用不同,部分呈现双相调节作用,而上市对照制剂消癌平注射液对IL-2主要表现为抑制作用而对TNF—α的影响不显著。结论抗肿瘤中药通光藤具有确切抗肿瘤作用,作用机理主要不是通过其直接细胞毒活性,而是通过调节肿瘤细胞的免疫功能起关键作用,通光藤是一种很有前途的抗肿瘤中药。

Abstract:Objective To study on the anti-tumour actions and their mechanisms of the extraetives from Marsdenia tenacissima. Methods Existing listed preparation Xiaoaiping injection was as positive control and conidia of Pyricularia oryzae and dual MTT method in vitro were used to screen anti-tumor bioactive constituents from Marsdenia tenacissima. Mice model of Hep 1 - 6 hepatocellular carcinoma was used to validate the in vivo active site of its inhibitory tumor efficacy. ELISA was furtherly tested to verify the effect of total glucoside extract on tumor necrosis factor-α(TNF-α) released by mice intra-abdominal maerophage and interleukin 2 (IL-2) produced by mice splenic cell. Results The direct cytotoxic activity of Marsdenia tenacissima was not obvious according to the results of double in vitro activity screening. The extracts had obvious anti-tumor function and some certain toxicity. There was a certain degree of influence on the release of TNF-αand IL-2 by either the chemical parts of Marsdenia tenaeissima or the positive control. Different chemical parts showed different effects and some of them present dual phase adjust action. Xiaoaiping injection showed the inhibitory action on IL-2 and no obvious influence on TNF-α. Conclusions Marsdenia tenacissima has definite anti-tumor functions by regulating the immune function of tumor cells and not mainly by the direct cytotoxic activity. It is a promising anti-tumor medicine.
