人子宫内膜腺上皮细胞及基质细胞分离方法的研究

摘    要:目的 探索子宫内膜腺上皮细胞及基质细胞分离的新方法。方法 通过胶原酶阶梯消化(0.25%,0.15%,0.1%,0.08%)、不同目次的筛网过滤、重力沉降等技术得到高纯度的人子宫内膜腺上皮细胞和基质细胞。结果 应用浓度逐渐降低的胶原酶消化子宫内膜组织,可提高细胞的存活率及细胞收集量。筛网过滤和重力沉降法,可提高子宫内膜腺上皮细胞和基质细胞的纯度,腺上皮细胞可达90%,基质细胞可达96%以上。结论 采用此方法可得到大量高纯度的子宫内膜腺上皮细胞及基质细胞,为体外研究子宫内膜提供模型。

Abstract:Objective To explore the new methods of isolating and culturing glandular epithelial cells and stromal cells of human endometrium.Methods Using the technique of different gradient(0 25%,0 15%,0 1%,0 08%)of collagenase,serial filtration and sedimentation,the higher purities of glandular epithelial cells and stromal cells were obtained.Results The yieds and the surviving rates of two kinds of cells were increased by collagenase-ladder digest assay.The purities of glandular epithelial cells and stromal cells enhanced,90% and 95% respectively,by serial filtration and sedimentation.Conclusions The high purities of glandular epithelial cells and the stromal cells can be used as one of the cell models in vitro by those techniques.
