银杏叶提取物EGb761对大鼠脑缺血再灌注后细胞凋亡及Bcl-2、Bax蛋白表达的影响

摘    要:目的:探讨EGb761对大鼠局灶性脑缺血再灌注后细胞凋亡及Bcl-2和Bax蛋白表达的影响。方法:48只SD雄性大鼠随机分为假手术组、模型组、EGb761低剂量组及EGb761高剂量组,每组12只。应用线栓法制备大鼠大脑中动脉缺血模型,TTC染色法测定脑梗死体积,应用TUNEL法检测神经元凋亡,应用Western blot方法检测Bcl-2及Bax蛋白表达。结果:与模型组比较,EGb761高低剂量组脑梗死体积显著减少(P<0.05),神经元凋亡数显著减少(P<0.01),Bax蛋白表达明显减少(P<0.05,P<0.01),Bcl-2/Bax比值明显升高(P<0.01);EGb761高剂量组Bcl-2蛋白表达明显增多(P<0.01)。EGb761高低剂量组间Bcl-2/Bax比值差异有统计学意义(P<0.05)。结论:EGb761能显著增加脑缺血再灌注后Bcl-2蛋白表达,减少Bax蛋白表达,抑制神经元凋亡,减少脑梗死体积,对脑缺血再灌注损伤发挥保护作用。

Abstract:Objective: To investigate the effect of EGb761 on apoptosis and expression of Bcl-2/Bax proteins after ischemia-reperfusion injury in rats.Methods: Forty-eight male rats were randomized into sham operation group,model group,low-dose EGb761 group and high-dose EGb761 group,12 rats in each group.The middle cerebral artery ischemia models were established by thread embolism method;the volume of cerebral infarction was detected by TTC staining method;the neuronal apoptosis was analyzed by TUNEL method;the expressions of Bcl-2 and Bax were measured by Western blot method.Results: Compared with model group,the volume of cerebral infarction(P<0.05),the number of neuronal apoptosis(P<0.01) and expression of Bax(P<0.05,P<0.01) were all decreased significantly in the EGb761 groups,and the ratio of Bcl-2 to Bax was increased obviously(P<0.01) in the EGb761 groups;the expression of Bcl-2 protein in high-dose EGb761 group was increased(P<0.01).There was a significant difference in Bcl-2/Bax ratio between high-dose and low-dose EGb761 groups(P<0.05).Conclusion: EGb761 can increase the expression of Bcl-2 protein,decrease the expression of Bax protein,restrain the neuronal apoptosis and reduce the cerebral infarction volume to protect the ischemia-reperfusion injury.
