艾灸百会穴对AD大鼠学习记忆能力及海马区CaMKⅡ mRNA表达的影响

摘    要:目的:观察艾灸百会穴对阿尔茨海默氏病(AD)大鼠学习记忆能力和海马区钙调蛋白激酶Ⅱ(CaMKⅡ)mRNA表达的影响。方法:用Morris水迷宫进行筛选建立AD大鼠模型。将符合模型标准的20只AD大鼠随机分模型组、艾灸组,每组10只。另外随机选取10只正常老年大鼠为对照组。艾灸组用特制艾条在百会穴施温和灸,共治疗6个疗程。治疗结束后进行Morris水迷宫测试大鼠学习记忆能力,并用RT-PCR法检测大鼠左侧海马组织CaMKⅡ表达水平。结果:模型组平均逃避潜伏期较对照组和艾灸组明显延长(P<0.05,P<0.01),有效区停留时间较对照组和艾灸组明显缩短(P<0.05,P<0.01);上述二指标艾灸组与对照组比较均无显著性差异(P>0.05)。三组之间平台停留次数无显著性差异(P>0.05)。模型组左侧海马区CaMKⅡmRNA相对表达量较对照组和艾灸组显著下降(P<0.05,P<0.01),艾灸组与对照组比较无显著性差异(P>0.05)。结论:艾灸百会穴可通过上调海马区CaMKⅡmRNA表达以提高AD大鼠学习记忆能力。

Abstract:Objective:To research the effects of moxibustion at Baihui(GV20) on learning and memory ability and calmodulin dependent protein kinase Ⅱ(CaMKⅡ) mRNA expression in Alzheimer's disease(AD) rats.Methods:AD model was established with Morris water maze experiment.Twenty AD rats were randomized into two groups:moxibustion group in which 10 AD rats were treated with moxibustion at Baihui(GV20) for six treatment courses and model group in which 10 AD rats were given no treatment;and another 10 old rats were enrolled as control group.The learning and memory ability of AD rats were measured by Morris water maze experiment,and the CaMKⅡ mRNA expression in the left hippocampus was detected with RT-PCR.Results:The platform locating latency(PLL) was longer in model group than in control group and moxibustion group(P〈0.05,P〈0.01),and the stay time in coverage area was shorter in model group than in control group and moxibustion group(P〈0.05,P〈0.01),but there was no difference in platform locating latency and stay time in coverage area between moxibustion group and control group(P〉0.05);no difference was found in the number of crossing platform between the three groups(P〉0.05).The CaMKⅡ mRNA expression was significantly lower in model group than in control group and moxibustion group(P〈0.05,P〈0.01),with no difference between moxibustion group and control group(P〉0.05).Conclusion:Moxibustion at Baihui(GV20) can up-regulate CaMKⅡmRNA expression to improve the learning and memory ability of AD rats.
