血清对U937细胞分化过程中p21WAF1蛋白表达的影响

摘    要:目的:探讨U937细胞分化过程中血清对p21WAF1蛋白表达的影响。方法:利用乙酸肉豆蔻佛波醇(PMA)诱导U937细胞分化,血清作为调节因素;实验分为血清组(FCS)、血清加PMA组(FCS+PMA)、无血清组(NOFCS)和无血清加PMA组(NOFCS+PMA),将血清组作为对照组;细胞的贴壁功能作为分化标志;Western印迹测定各组p21WAF1蛋白表达。结果:PMA作用U937 24 h后,FCS+PMA组细胞贴壁生长,而其他各组细胞悬浮生长;FCS+PMA组细胞贴壁率明显高于对照组,差异有显著性(P<0.01),而NOFCS和NOFCS+PMA组细胞贴壁率与对照组相比无明显差异(P>0.05)。FCS+PMA组和NOFCS+PMA组与FCS组相比p21WAF1蛋白呈高度表达,差异有显著性 (P<0.05);而NOFCS组与FCS组相比p21WAF1蛋白表达差异无显著性 (P>0.05)。 结论:血清不影响PMA作用U937细胞内p21WAF1蛋白表达,但影响细胞分化成熟。

Abstract:ObjectiveTo study the effects of serum on p21 WAF1 expression during U937 cell differentiation. MethodsU937 cell differentiation induced by PMA (Phorbol Ester) was used and divided into four experiment groups according to whether or not PMA and serum exsited in culture system: serum group (FCS), serum plus PMA group(FCS+PMA), serum free group(NOFCS), serum free plus PMA group (NOFCS+PMA). FCS group was used as control group. Cell adherence was identified as differentiation characteristics,p21 WAF1 protein expression was determined by Western Blot. ResultsAfter PMA was used for 24 hours, cell stick growth ,U937 cell adherence and p21 WAF1 protein expression in FCS+PMA group increased significantly compared with control group. Cell suspend growth and U937 cell adherence in NOFCS and NOFCS+PMA groups were no significant difference compared with control group. p21 WAF1 protein expression in NOFCS+PMA group increased significantly, but that in NOFCS had no significant difference compared with control group. ConclusionSerum can′t affect p21 WAF1 expression during U937 cell differentiation induced by PMA,but it can affect cell differentiaiton maturation.
