山楂叶总黄酮对脑缺血的保护作用

摘    要:目的:探讨山楂叶总黄酮(FMCL)对脑缺血的保护作用。方法:采用线栓法制作大鼠局灶性脑缺血模型(MCAO),并对其进行行为指标评分,检测其血清中超氧化物歧化酶(SOD)、肌酸激酶(CK)、乳酸脱氢酶(LDH)活性及丙二醛(MDA)的含量。采用血瘀法制作大鼠血瘀模型并检测其全血黏度(ηB)、血浆黏度(ηP)及血小板最大聚集率(MPAG),观察FMCL对血黏度及血小板聚集率的影响。结果:与模型组比较,FMCL 60和30 mg·kg-1可明显升高MCAO大鼠血清中SOD活性,降低血清中CK、LDH活性及MDA含量(P<0.05); FMCL 60和30 mg·kg-1可明显降低血瘀大鼠的全血黏度、血浆黏度及血小板聚集率(P<0.01,P<0.05)。结论:山楂叶总黄酮可减轻脑缺血损伤,对脑细胞具有保护作用,其机制可能与降低全血黏度、抑制血小板聚集有关。

Abstract:Objective To study the protective effects of flavone mixture of crataegus leaves(FMCL) on cerebral ischemia.Methods The local cerebral ischemia model was initiated by the middle cerebral artery occlusion(MCAO) in rats for 4 h.The neurologic deficit score and the activities of creatine kinase(CK),lactata dehydrogenase (LDH),superoxide(SOD) and the content of serum malondialdehyde(MDA)in MCAO rats were measured.At the same time blood stasis models were built in rats. The effects of FMCL on platelet aggregation,blood viscosity(ηB) and plasma viscosity(ηP) were detected.Results Compared with the model group,FMCL(60 and 30 mg·kg~(-1)) increased SOD activity,decreased CK and LDH activities and serum MDA content in a dose dependent manner in MCAO rats(P<0.05).It could also decrease the neurologic deficit in MCAO rats(P<0.05).And it could decrease platelet aggregation rate,ηB and ηP(P<0.01,P<0.05).Conclusion FMCL has protective effects on cerebral ischemia and its mechanism may be related to lowing ηB,ηP and preventing platelet aggregation.
