非Ca2+依赖型磷脂酶A2基因单核苷酸多态性与2型糖尿病发病的关联性分析

摘    要:目的: 探讨非Ca2+依赖型磷脂酶A2(iPLA2)基因家族中的PLA2G6基因单核苷酸多态性(SNPs)与中国北方汉族人群中2型糖尿病(T2DM)发病的关联性,阐明PLA2G6基因是否为T2DM的易感基因。 方法: 选择168例T2DM患者(病例组)和199名正常对照者(对照组),采用聚合酶链式反应(PCR)技术和限制性片段长度多态性(RFLP)的方法检测2组研究对象PLA2G6基因rs2235346、 rs2272831和 rs2284060的基因型及等位基因频数分布。 结果: PLA2G6基因rs2235346、 rs2272831和 rs2284060位点在病例组和对照组中的基因型频数分布均符合Hardy-Weinberg 平衡定律(P>0.05);PLA2G6基因rs2235346、 rs2272831和 rs2284060位点的基因型及等位基因频数在病例组和对照组分布比较差异均无统计学意义(χ2=0.728,P>0.05)。 结论: PLA2G6基因rs2235346、 rs2272831和 rs2284060位点多态性与T2DM的发病可能无关联,PLA2G6基因可能不是T2DM的易感基因。

Abstract:Objective: To detect the association between the single nucleotide polymorphisms(SNPs)of PLA2G6 gene of Ca2+-independent phospholipase A2(iPLA2)gene family and the incidence of type 2 diabetes mellitus(T2DM)in Chinese northern Han people,and to discuss whether the PLA2G6 gene was the susceptible gene of T2DM.Methods: 169 patients with T2DM were selected as case group and 199 healthy controls without T2DM were selected as control group.The techniques of polymerase chain reaction (PCR)and restriction fragment length polymorphism (RFLP)were used to examine the genotypic and allic frenquencies of rs2235346,rs2272831,and rs2284060 sites at PLA2G6 gene of the objects in case and control groups.Results: The distribution of genotypic and allic frequencies of rs2235346,rs2272831,and rs2284060 sites at PLA2G6 gene did not deviate from Hardy-Weinberg equilibrium in both case and control groups (P>0.05).The distribution of genotypic and allic frequencies of rs2235346,rs2272831,and rs2284060 sites at PLA2G6 gene had no significant differences between case and control groups(χ2=0.604,P>0.05).Conclusion: There is no significant relationship between the genotypic and allic frequencies of rs2235346,rs2272831,rs2284060 sites at PLA2G6 gene and T2DM,and the PLA2G6 gene is not the susceptible gene of T2DM.
