摘    要:目的:研究外源性p16基因对人前列腺癌PC-3M细胞系生长及致瘤性的抑制作用,探索前列腺癌基因治疗的新途径.方法:利用携带约800 bp的外源性p16 cDNA的真核表达质粒,通过脂质体介导将其导入人前列腺癌PC-3M细胞系中,用G418筛选阳性克隆,经流式细胞仪及细胞生长曲线测定等方法研究p16基因对人前列腺癌细胞周期生长特性的影响,并观察导入外源性p16基因的人前列腺癌细胞PC-3M对裸鼠致瘤能力的影响.结果:野生型p16基因转染到有p16基因缺失的人前列腺癌细胞PC-3M上,细胞周期明显变化,细胞从G1期到S期发生抑制,细胞增殖活性降低.结论:转染外源性p16基因可阻止人前列腺癌细胞由G1期进入S期,并使肿瘤恶性增殖受到抑制.

Abstract:Objective: To investigate the suppressive effect of exogenous p16 on the human prostate cancer PC 3M cell line and to probe a new approach to gene therapy of prostate cancer. Methods: A eukaryotic expression vector pcDNA3 and pcDNA3 p16 was transfected, respectively, into PC 3M cells by lipofectin. After positive clone was selected by G418, cell growth properties and cell cycle pattern were assessed by flow cytometry. Effect of p16 on growth cycle of PC 3M cell and the effect of PC 3M p16 cell on tumorgenicity in nude mice was examined. Results: The progression of the transfected PC 3M cells cell cycle from G 1 to S phase was arrested. The growth of cells transfected with p16 cDNA in nude mice was dramatically inhibited in comparision with the parent cells. Conclusion: Human prostate cancer PC 3M cell line could be arrested at G 1/S phase and the growth of cells could be significantly suppressed by exogenous gene p16 cDNA.
