基于数据挖掘筛选植物类抗肝癌中药及抗肝癌组方思路探讨

摘    要:通过检索2015年版《中华人民共和国药典(一部)》、《中药学》以及PubMed、中国知网、维普等国内外数据库,对植物类抗肝癌中药的品种进行挖掘整理,对其抗瘤机制以及性味归经等进行丰度分析,并结合肿瘤分期进行植物类抗肝癌中药组方设计探索。结果共检索得到具有确切抗肝癌活性的植物类中药120味(如桑枝、枸杞子、秦艽、太子参等),其抗瘤机制以抑制肿瘤增殖及生长、调节免疫、诱导凋亡为主,其丰度分别为0.783、0.700、0.467;药性以寒、温、平为主,丰度分别为0.408、0.283、0.183;药味以苦、甘、辛为主,丰度分别为0.625、0.525、0.308;归经以肝、肾、肺、胃、脾经为主,丰度分别为0.663、0.358、0.325、0.317、0.275。抗肝癌植物类中药组方思路如下:(1)肝癌早期宜以抑制肿瘤增殖及生长为主,可选择性寒味苦辛归肝经的植物类中药;(2)肝癌中期宜以抑制肿瘤侵袭及转移为主,可选择性平味甘归胃、脾经的植物类中药;(3)肝癌晚期宜以调节免疫和诱导凋亡为主,可选择性温平味苦甘归肾、肺经的植物类中药;(4)治疗过程中同时辅以具肠道微环境调节作用的植物类中药。结合中医辨证和患者病情特征进行个体化组方,以充分发挥中药对肝癌的治疗作用。研究结果可为临床抗肝癌中药组方及新药研发提供参考。

Screening of Anti-tumor Botanical Chinese Medicine for Liver Cancer Based on Data Mining and Exploration of Anti-tumor Botanical Chinese Medicine Formula for Liver Cancer

Abstract:By retrieving from Chinese Pharmacopeia(part 1;2015 edition),Chinese Materia Medica,and the databases of PubMed,CNKI and VIP,we mined and summarized the varieties of anti-tumor botanical Chinese medicine for liver-cancer,and analyzed the abundance of their anti-tumor mechanism,property,flavor,and meridian tropism.The design for botanical Chinese medicine formulae in treating liver cancer according to tumor staging was also explored.The results showed that a total of 120 herbs such as Ramulus Mori,Fructus Lycii,Radix Gentianae Macrophyllae and Radix Pseudostellariae which have certain anti-tumor activities were obtained.The anti-tumor mechanism of the 120 kinds of botanical Chinese medicine mainly manifested as inhibition of tumor proliferation and growth,regulation of immunity,and induction of apoptosis,with the abundance of 0.783,0.700,0.467,respectively.The main properties of the 120 kinds of botanical Chinese medicine were cold,warm and mild,with the abundance of 0.408,0.283,0.183,respectively.The main flavors of the 120 kinds of botanical Chinese medicine were bitter,sweet and spicy,with the abundance of 0.625,0.525 and 0.308,respectively.The main meridian-tropisms of the 120 kinds of botanical Chinese medicine were with tropism to the meridians of liver,kidney,lung,stomach and spleen,with the abundance of 0.663,0.358,0.325,0.317,0.275,respectively.The thoughts for botanical Chinese medicine formula in treating liver cancer were as follows:(1)for early stage of liver cancer,the therapeutic target is to inhibit tumor proliferation and growth,and the cold,bitter and spicy herbs with the meridian tropism to liver meridian can be used;(2)for middle stage of liver cancer,the therapeutic target is to inhibit tumor invasion and metastasis,and the mild and sweet herbs with the meridian tropism to stomach and spleen meridians can be used;(3)for late stage of liver cancer,the therapeutic target is to regulate immunity and induce apoptosis,and the warm,mild,bitter and sweet herbs with the meridian tropism to kidney and lung meridians can be used;(4)additionally,herbs with the actions of regulating intestinal microenvironment can be used for adjuvant therapy during the treatment.The individualized formula based on syndrome differentiation and the illness state of the patients will enhance the therapeutic effect on liver cancer.The results will supply evidence for the research and development of new formulae and new drugs for the treatment of liver cancer.
