ARMCX1和MMP-9在宫颈鳞癌中的表达研究

摘    要:目的 检测宫颈鳞癌组织中ARM重复X连锁蛋白1(ARMCX1)和基质金属蛋白酶-9(MMP-9)的表达特征,分析二者在不同临床病理特征中的表达差别,探讨二者的相关性及对预后判断的价值。方法 本实验以67例宫颈鳞癌的患者临床资料及留取的术后组织作为观察组,以67例非肿瘤性宫颈组织作为对照组,应用免疫组化方法、流式细胞术及Western Blot检测二组中ARMCX1和MMP-9的表达。结果 免疫组化结果显示ARMCX1蛋白主要定位于细胞浆和细胞膜中,少数病例定位于细胞核;MMP-9蛋白定位于细胞浆和细胞膜。流式细胞结果显示二者的表达量在二组间的差别有统计学意义,观察组中ARMCX1和MMP-9的表达在不同脉管累犯、病变分型及淋巴结转移的分组中差别有统计学意义。ARMCX1的表达与在不同增殖指数的分组中差别有统计学意义。ARMCX1和MMP-9具有正相关性。ARMCX1和MMP-9的表达与患者的预后有关。结论 宫颈鳞癌中ARMCX1和MMP-9表达升高,且具有正相关性,对肿瘤形成和进展有促进作用。联合检测二者的表达对判断预后有重要意义。

Expression study of ARMCX1 and MMP-9 in cervical squamous cell carcinoma
Abstract:Objective To detect the expressions of ARMCX1 and MMP-9 in cervical squamous cell carcinoma tissues, analyze difference of their clinicopathological features, and to explore their correlation and prognostic value. Methods 67 patients with cervical squamous cell carcinoma were selected as observation group, 67 non-neoplastic cervical tissues were selected as control group. The expressions of ARMCX1 and MMP-9 were detected by immunohistochemistry, flow cytometry and Western Blot method in two groups. Results IHC results showed that ARMCX1 protein was mainly localized in cytoplasm and cell membrane, and a few cases were localized in nucleus. MMP-9 protein was localized in cytoplasm and cell membrane. Flow cytometry showed that the expression of ARMCX1 and MMP-9 was significantly different in two groups. The expressions of ARMCX1 and MMP-9 were closely related to vascular involvement, pathological classification and lymph node metastasis in the observation group. The expression of ARMCX1 was closely related to the proliferation index of tumors. Positive correlation were found between ARMCX1 and MMP-9 by correlation analysis in the observation group. Expressions of ARMCX1 and MMP-9 were related to prognosis. Conclusion The expressions of ARMCX1 and MMP-9 is significantly increased, has a synergistic and positive effect in cervical squamous cell carcinoma. ARMCX1 and MMP-9 can regulate the formation and development of cervical squamous cell carcinoma. ARMCX1 and MMP-9 may be value to judge prognosis.
