HMGB1和RAGE在口腔扁平苔藓中的表达及意义

摘    要:目的 探讨高迁移率转移族蛋白(HMGB1)和晚期糖基化终末产物受体(RAGE)在口腔扁平苔藓(OLP)中的表达及意义.方法 选择经病理确诊的OLP患者和健康体检者各30例,采用免疫组化法观察HMGB1及RAGE蛋白在其口腔黏膜组织中的表达情况;运用实时荧光定量PCR技术检测HMGB1及RAGEmRNA在其外周血中的表达量.结果 在扁平苔藓患者的口腔黏膜组织中,HMGB1和RAGE的蛋白表达水平均高于健康对照组(P<0.05).HMGB1与RAGE的蛋白表达水平呈正相关性(r=0.473,P<0.05);在口腔扁平苔藓患者外周血中,HMGB1和RAGE的基因表达水平均高于健康对照组(P<0.05);HMGB1与RAGE的基因表达水平呈正相关性(r =0.922,P<0.05).结论 HMGB1和RAGE协同参与了OLP的发病过程,其机制有待于进一步研究.

Abstract:Objective To investigate the expression and significance of HMGB1 and RAGE in the development of oral lichen planus.Methods 30 cases with OLP and 30 healthy controls were selected.The protein expression of HMGB1 and RAGE in the oral mucosa were examined with immunohistochemical technique.HMGB1 and RAGE mRNA expression in the peripheral blood were detected by the real-time fluorescent quantitative PCR.Results The expression level of HMGB1 and RAGE in oral lichen planus was higher than that in control group (P < 0.05).The positive expressing rates of HMGB1 and RAGE in oral lichen planus were 86% and 70%,respectively.In the patients group with oral lichen planus,the expression level of HMGB1 and RAGE mRNA showed a positive correlation (r =0.922,P < 0.05).The protein expression level of HMGB1 and RAGE also showed a positived correlation (r =0.473,P < 0.05).Conclusion RAGE and HMGB1 have correlations in the pathogenesis of OLP.The mechanism remains to be further probed.
