育阴灵冲剂对自然流产小鼠蜕膜细胞凋亡调控基因作用的实验研究

摘    要:目的探讨补肾中药方育阴灵冲剂对自然流产小鼠的保胎机理。方法建立正常妊娠模型CBA/J×BALB/c和自然流产模型CBA/J×DBA/2。正常妊娠模型孕鼠灌服生理盐水,自然流产模型孕鼠随机分为中药组(灌服育阴灵冲剂)、西药组(腹腔注射CsA)、模型组(灌服生理盐水)。孕13 d观察各组小鼠的流产情况,用免疫组化测定4组孕鼠蜕膜细胞Bcl-2、Bax、Fas、FasL蛋白的表达,并对其染色强度进行半定量分析。结果中药组、西药组胚胎吸收率明显低于模型组(P0.05),与正常组相比,中药组、西药组的胚胎吸收率变化不明显(P0.05)。与模型组相比,中药组、西药组小鼠蜕膜细胞FasL、Bax的表达降低,Bcl-2表达升高(P0.05);与正常组相比,中药组、西药组FasL的表达均有所升高(P0.05),Bax的表达升高,Bcl-2的表达降低(P0.05);中药组与西药组比FasL、Bax、Bcl-2的表达均无统计学意义(P0.05);Fas的表达各组比较无明显差异(P0.05)。结论育阴灵冲剂的保胎作用可能与其纠正免疫失衡状态及降低流产率有关。

Abstract:Objective To explore the mechanism of yuyinling infusion on fetus protection in mice with spontaneous abortion. Methods Normal pregnant model CBA/J × BALB/c and spontaneous abortion model CBA/J × DBA/2 were established. Normal saline was administered in normal pregnant models. The spontaneous abortion models were divided into Chinese medicine group ( administration of yuyinling infusion) , western medicine group( intraperitoneal injection of CsA) , and model group( administration of normal saline). Abortion was observed in 13 days of pregnancy in each group. HIC was adopted to determine the expressions of Bcl - 2, Bax, and Fas, FasL proteins in deeidua cells in mice of four groups. Semi - quantitative analysis was applied to determine the staining strength. Results Embryo absorption in Chinese medicine and western medicine groups were lower apparently than that in model group( P 〈 0. 05 ). Compared with normal group, in Chinese medicine and western medicine groups, the expressions of FasL and Bax in deeidua cells decreased and the expression of Bcl-2 increased(P 〈 0. 05). Compared with normal group, the expression of FasL in Chinese medicine and western medicine groups increased( P 〉 0.05 ) , that of Bax increased( P 〈 0.05 ) and that of Bcl - 2 decreased. In comparison between Chinese medicine and western medicine groups, the statistical significance did not present in the expressions of FasL, Bax and Bcl -2 (P 〉 0. 05). There was no apparent differenee in Fas expression in comparison of four groups(P 〉 0.05). Conclusion Yuyinling infusion presents the protection on fetus, which is probably relevant with its rectification of immune imbalance and decrease of abortion rate.
