摘    要:目的观察不同电针"百会""印堂"对抑郁模型大鼠结肠黏膜三种胃肠道相关脑肠肽胃泌素(GAS)、神经肽Y(NPY)及降钙素基因相关肽(CGRP)的影响,为揭示针刺治疗抑郁症胃肠道躯体化症状作用途径提供实验依据,并比较不同电针的作用效果。方法将60只健康SD大鼠随机分为空白组、模型组、百优解组、脉冲电针组、音乐电针组,每组12只。采用慢性不可预见的温和刺激结合孤养方法制备抑郁模型。百忧解组每天将百忧解以生理盐水配制灌胃给药。脉冲电针组针刺大鼠"百会"和"印堂"穴,接脉冲电针治疗仪。音乐电针组取穴同脉冲点针组,接音乐电针治疗仪。采用放射免疫法测定各组大鼠结肠黏膜GAS、NPY、CGRP含量。结果模型组与空白组相比,大鼠结肠黏膜GAS含量显著降低(P<0.05),NPY含量显著升高(P<0.05),CGRP含量极显著升高(P<0.01);脉冲电针组、音乐电针组及百忧解组与模型组相比,大鼠结肠黏膜GAS含量显著升高(P<0.05或P<0.01),NPY含量显著降低(P<0.05或P<0.01),CGRP含量显著降低(P<0.05或P<0.01);音乐电针组与脉冲电针组相比,大鼠结肠黏膜CGRP含量降低水平更为显著(P<0.05)。结论电针治疗可通过调整抑郁后胃肠道兴奋性激素与抑制性激素的失平衡来影响肠道中脑肠肽含量,从而调控脑-肠轴,使抑郁后胃肠功能紊乱状态得到改善,而且音乐电针组优于脉冲电针组。

Impacts on the Contents of Brain - Gut Peptides ( GAS, NPY and CGRP) in Colonic Mucosa of Depression Model Rats Treated with Different Electroa- cupuncture

Abstract:Objective To observe the impacts on three gastrointestinal tract related brain - gut pep- tides, named gastrin ( GAS ) , neuropeptide Y ( NPY ) and calcitonin gene related peptide ( CGRP ) in colonic mucosa treated with electroaeupuneture (EA) at Baihui ( GV20 ) and Yintang (Extra) in depression model rats so as to discover the experimental evidences for acupuncture effective approach in the treatment of gastroin- testinal somatization symptoms of depression and make the comparison of the effects among different EA ther- apies. Methods Sixty healthy SD rats were randomized into a blank group, a model group, a Prozac group, a pulse EA group and a music EA group, 12 rats in each one. Depression model was prepared by chronic unpre- dictable mild stimulation and isolated raising method. In the Prozac group, Prozac was prepared with physical saline for gastric infusion. In the pulse EA group, acupuncture was applied at Baihui ( GV20 ) and Yintang (Extra) ,attached by pulse electric acupuncture apparatus. In the music EA group,the points were same as the pulse EA group, but attached by the music electric acupuncture apparatus. The radioimmunoassay was a- dopted to detect the contents of GAS, NPS and CGRP of colonic mucosa of the rats in each group. Results Compared with the blank group, GAS content was reduced significantly ( P 〈 0.05 ) , NPY was increased sig- nificantly( P 〈0.05 )and CGRP was increased the most significantly (P 〈 0.01 )in the model group. Com- pared with the model group,GAS content was increased significantly(P 〈0.05 or P 〈0.01 ) ,NPY was re- duced significantly ( P 〈 0.05 or P 〈 0.01 ) and CGRP was reduced significantly( P 〈 0.05 or P 〈 0.01 ) in the pulse EA group, music EA group and Prozac group. CGRP content in the music EA group was reduced muchmore significantly as compared with the pulse EA group ( P 〈 0.05 ). Conclusion Electroacupuncture im- pacts the contents of brain - gut peptides in the intestinal tract through regulating the imbalance between gas- trointestinal excitatory hormone and inhibitory hormone after depression so as to control and adjust brain - in- testine axis and improve in gastrointestinal disturbance. The results in the music EA group is superior to the pulse EA group.
