原发性干燥综合征患者T细胞亚群及CD4~+CD25~+ T细胞检测的意义

摘    要:目的探讨T淋巴细胞亚群及CD4+CD25+T细胞在原发性干燥综合征(PSS)患者中的检测意义。方法用流式细胞仪检测18例PSS患者以及20例健康体检者外周血CD3+、CD3+CD4+、CD3+CD8+、NK细胞(CD3-/CD16+56+)、CD3+CD4-CD8-双阴性T细胞及CD4+CD25+调节T细胞水平。结果 PSS患者与正常对照组比较,CD3+CD4+细胞亚群的百分率明显升高(P<0.05);CD4+/CD8+与对照组比较有明显升高(P<0.05);CD3+CD4-CD8-双阴性T细胞及CD4+CD25+调节性T细胞的百分率明显降低(P<0.01),其它T细胞亚群无明显变化。结论 PSS患者T淋巴细胞亚群的水平是异常的。T细胞亚群的比例异常与疾病的病程和临床表现相关联。CD3+CD4-CD8-和CD4+CD25+调节性T细胞水平降低导致机体自身免疫反应抑制功能减弱,免疫耐受状态被打破,可能是引发PSS发生发展的重要因素。

Abstract:Objective To investigate the significance of T lymphocyte subsets and CD4+CD25+ T cells detection in patients with primary Sjogren syndrome(PSS).Methods A flow cytometry was use to detect the levels of peripheral blood CD3+,CD3+CD4+,CD3+CD8+,CD3-/CD16+56+ of NK cells,double negative T cells of CD3+CD4-CD8-and CD4+CD25+ which regulate the levels of T cells in 18 patients with PSS,and compared with 20 healthy subjects(as normal control group).Results Compared with normal control group,the percentage of PSS patients' CD3+CD4+cell subsets were significantly elevated(P<0.05);CD4+/CD8+ was also significantly higher(P<0.05); double negative T cells of CD3+CD4-CD8-and CD4+CD25+ which regulate the levels of T cells decreased significantly(P<0.01),other T cell subsets remained the same.Conclusion The levels of T lymphocyte subsets in patients with PSS is abnormal.The proportion of T cell subsets abnormalities was associated with the course and clinical manifestations of the disease.The decreased CD3+CD4-CD8-and CD4+CD25+ which regulate the levels of T cells resulted in weakening self immune suppression,and immune tolerance has been broken,they may be important factors which induced the genesis and development of PSS.
