FEZ1/LZTS1基因与原发性肝癌相关性研究

目的:研究FEZ1/LZTS1基因和原发性肝癌的相关性。方法:新鲜肝癌组织及正常肝脏组织各96例,分别提取癌组织(其中肝细胞癌86例,胆管上皮癌6例,混合性癌4例)和正常组织总RNA,采用RT-PCR方法检测肝癌组织和正常肝脏组织FEZ1/LZTSl基因mRNA的表达。运用免疫组织化学法(SP法)分别检测96例肝癌和正常肝脏组织标本FEZI/LZTSl基因蛋白表达。结果:FEZ1/LZTS1基因的mRNA表达在96例肝癌组织中的情况如下:27例(28.1%)缺失,28例(29.2%)表达下降,41例(42.7%)表达正常;而FEZ1/LZTS1基因mRNA在相应的96例正常肝脏组织中全部表达。肝癌组织FEZ1/LZTS1基因的mRNA表达与正常肝脏组织相比显著下调(P<0.01)。FEZ1/LZTS1基因蛋白在全部正常肝脏组织中表达,在肝癌组织中30例(31.3%)强阳性(+),20例(20.8%)中度阳性(+/-),26例(27.1%)弱阳性(-/+),20例(20.8%)阴性(-)。结论:与正常组织比,FEZ1/LZTS1基因和蛋白表达在原发性肝癌组织中明显缺失或减少,这可能导致肝癌的发生。

Correlation between FEZ1/LZTS1 gene and primary hepatocellular carcinoma

Objective: To explore the correlation between FEZ1/LZTS1 gene and primary hepatocellular carcinoma.Methods:Expression of FEZ1/LZTS1 gene mRNA in liver cancer.Total RNA of 96 cases of liver cancer(86 cases with hepatocellular carcinoma,6 cases with bile duct epithelioma,and 4 cases with mixed carcinoma) and 96 cases of normal liver tissues were extracted,and the mRNA expression of FEZ1/LZTS1 gene was detected by RT-PCR.The expression of FEZ l/LZTS l protein was detected in 96 cases paraffin embedded specimen using immnunohistochemical assay.Results : The expression of FEZ1/LZTS1 gene mRNA of 96 cases liver cancer tissues was as follow: 27 cases(28.1%) were absent,28 cases(29.2%) were down regulated,and 41 cases(42.7%) were normally expressed.Accordingly,all 96 normal liver tissues had the mRNA expression.The difference between carcinoma group and normal group was statistically significance(P<0.01).FEZ l/LZTS l protein was expressed in normal liver tissues.In 96 cases of liver cancer,the FEZ1/ LZTSl protein expression was as following: 20 cases(+/-),26 cases(-/+),20 cases(-),other 30 cases(+).Conclusions: Absence or down regulation of FEZ1/LZTS1 gene and protein may cause liver cancer.
