摘    要:目的:酵母Nar1p是人类早老症关键蛋白prelamin A相互作用蛋白NARF在酵母中的同源蛋白,目前未见其在寿命和衰老中的功能报道。本研究以酿酒酵母为模式物种,研究Nar1p在酵母复制寿命和氧化应激中的作用。方法:首先通过基因重组的原理构建Nar1p过表达酵母菌株,然后检测其复制寿命、对强氧化剂百草枯的抵抗能力、细胞内总SOD活性。结果:相对于野生型酵母菌株,Nar1p过表达酵母菌株的复制寿命缩短,抗氧化性损伤的能力减弱,但是细胞内总SOD活性升高。结论:Nar1p很可能参与了酵母复制寿命和氧化应激能力的调节,为全面研究Nar1p的功能奠定基础。

Abstract:Objective: Saccharomyces cerevisiae Nar1p is the orthologue protein of human NARF. NARF binds specifically to prenylated prelamin A, which is involved in Hutchinson-Gilford progeria syn drome, a dominant autosomal premature aging syndrome. But the function of NARF has not been repor ted. Using Saccharomyces cerevisiae as the model, the study aimed to investigate Nar1p function in lifes pan regulation and its antioxidantive ability. Methods: Nar1p overexpression yeast strain was made by ge- nome homologous recombination to analyze the replicative lifespan, its antioxidative ability to paraquat and total SOD activity. Results: Nar1p overexpression yeast strain showed reduced replicative lifespan, de- creased antioxidative ability but upregulated total SOD activity. Conclusion: Nar1p maybe involved in rep- licative lifespan and antioxidative ability regulation.
