摘    要:【目的】用小剂量地塞米松诱发大鼠出现胰岛素抵抗,建立胰岛素抵抗动物模型,并对模型的稳定性进行初步评价。【方法】大鼠4组,给药组皮下注射20、60、120μg/kg 3种不同剂量地塞米松溶液,2次/d,对照组皮下注射等量的生理盐水。【结果】20μg/kg组大鼠在21 d时空腹血糖升高,胰岛素耐量实验结果表明其存在胰岛素抵抗,体重只有轻度下降,停药7 d后,胰岛素敏感性从抵抗状态恢复到正常水平。【结论】小剂量的地塞米松连续皮下注射将诱发大鼠出现一过性的胰岛素抵抗,模型动物的一般状态好。

Abstract:Objective] To establish a new animal model for the study of insulin resistance. Methods] Different dosage (20, 60, 120μg/kg) of dexamethasone were given to wistar rat (8 or 12 pergroup) for 28 d to indace the resistance for insulin. Fasting blood glucose and insulin sensitivity were determined. Results] Fasting blood glucose and insulin sensitivity were significantly reduced by 20 μg/kg dexamethasone on 21 d, and with slight decrease in body weight as compared with vehicle. After dexamethasone was withdrawn for 7 d, fasting blood glucose and insulin sensitivity recovered. Conclusions] Chronic low-dose subcutaneous dexamethasone (20μg/day) may cause a transient model for the study of insulin resistance.
