摘    要:【目的】改进大鼠心肌缺血再灌注动物模型制作方法。【方法】雄性Sprague-Dawley大鼠麻醉开胸后,用310医用单丝线穿过冠状动脉,在结扎线下垫聚乙烯管,结扎缝线缺血30min,剪断缝线再灌注120min。可省却呼吸机。【结果】24只SD大鼠,行单纯冠状动脉结扎术模型组大鼠存活率为10/12,再灌注后模型组存活率为9/12。假手术组存活率为12/12。经过氯化三苯基四氮唑(TIE)染色,梗死心肌和正常心肌组织分界清晰。Nagar Olsen心肌组织特殊染色,光镜下对照组心肌组织染成黄色,而模型组心肌组织可见大片红染的阳性反应区。【结论】采用本方法制作大鼠心肌缺血/再灌注损伤模型成功率较高,且操作简单、创伤小。

Abstract:Objective] To improve the procedure of estabhshing animal model of myocardial ischemia/reperfusion injury in rats. Methods] Male Sprague-Dawley rats were anesthetized, the chest was opened without respirator, a 3/0 silk ligature was placed around the left main coronary artery, a polyethylene tube was placed under the silk ligature. The coronary artery was occluded by tightening the ligature for 30 min, reperfusion was achieved by releasing the tension applied to the ligature for 120 min. Results] A total of 24 Sprague-Dawley rats were allocated into model group and control group. During 30 min of myocardial ischemia, 10/ 12 model group rats survived. Following 120 rain myocardial reperfusion 9/12 operated group rats survived. Conclusions] A simple and reliable technique for animal model of myocardial ischemia/repeffusion injury in rat has been developed without respirator, which may result in a higher success rate.
