Staurosporine对甲状腺癌SW579细胞的促凋亡作用

【目的】研究星形孢菌素促进人甲状腺癌细胞SW579凋亡的作用,从而为肿瘤治疗寻找新的方向。【方法】MTT法检测ST对SW579细胞增殖的抑制作用。流式细胞术检测细胞周期分布及细胞凋亡情况。用Hochest33258及MDC染色方法分别检测细胞的凋亡及自噬发生情况。Western blotting用于检测相关蛋白的表达情况。【结果】通过MTT检测发现staurosporine对SW579增殖的抑制呈浓度和时间依赖性。流式细胞术检测发现staurosporine对SW579细胞具有G0~G1期阻滞。【结论】Staurosporine能够诱导对SW579细胞凋亡从而抑制肿瘤细胞的增殖。

Effect of staurosporine on promoting apoptosis of cell line SW579 of human thyroid cancer

【Objective】To investigate the effects and mechanism of staurosporine on human thyroid cancer SW579 cell line in vitro.【Methods】Cell growth inhibition of SW579 induced by staurosporine was assessed with methylthiazolyl tetrazolium（MTT） assay,cell cycle distribution was evaluated by flow cytometry and apoptosis induction was also evaluated by flow cytometry.【Results】Methyl thiazolyl tetrazolium（MTT） assays indicated that staurosporine decreased the viability of SW579 cells in a dose-and time-dependent manner.Flow cytometry analysis indicated that staurosporine increased cell population at G0~G1 phase,an index of apoptosis.【Conclusion】Staurosporine can induce SW579 cell apotosis,thus inhibiting proliferation of tumor cell.
