TZC对SD复合DU污染大鼠SS神经元的保护作用

摘    要:【目的】探讨牛磺酸锌(taurine-zinc coordination compound,TZC)对贫铀(depleted Uranium,DU)污染复合睡眠剥夺(sleep deprivation,SD)大鼠皮层生长抑素(somatostatin,SS)阳性神经元的保护作用。【方法】Wistar大鼠贫铀颗粒气管灌注后3个月,采用小平台水环境法建立大鼠睡眠剥夺模型,观察大鼠睡眠剥夺72h后大脑皮层生长抑素阳性神经元变化情况,并以牛磺酸锌进行保护。【结果】光镜下NS组大鼠皮层SS阳性神经元均匀分布。NSSD组、DU组和DUSD组SS阳性神经元染色浅,尤以DUSD组为甚。DU+TZC组SS阳性神经元染色较DU组深,DUSD+TZC组与DUSD组比较也较深。DU组皮层SS阳性神经元数目比NS组明显减少(P〈0.05),NSSD与NS、DUSD组比DU组均明显减少(P〈0.05),DUSD+TZC组与DU+TZC组比较无明显差别(P〉0.05)。DUSD组阳性神经元数目比NSSD组明显减少(P〈0.05),而DUSD+TZC组与NSSD组比较仍无明显差别(P〉0.05)。【结论】贫铀污染能引起皮层SS阳性神经元形态改变和数目减少,SD可以使这种效应加剧。牛磺酸锌可能通过影响大脑皮层SS阳性神经元保护大鼠受损的脑功能。

Abstract:Objective] To explore the protective effects of taurine-zinc Coordination compound (TZC) on the somatostatin positive cortical neurons of rats after the effects of sleep deprivation (SD) and depleted uranium (DU) Methods] SD was induced in Wistar rats by employing flower pot 3 months after they were exposed to DU particles intratracheal instillation. Somatostatin positive cortical neurons of rats exposed to DU were compared after 72 h, so did that of rats fed with TZC. Results] Microscopic observertion showed that somatostatin positive cortical neurons of NS group distributed evenly. Staining density of NSSD, DU and DUSD were lesser gradually, especially in DUSD group. DU + TZC group was stronger than DU, while DUSD + TZC was stronger than DUSD. The number of somatostatin positive cortical neurons decreased in DU group as compared with NS group( P 〈 0.05). NSSD group was less than NS, while DUSD group was lesser than DU( P 〈 0.05). There was no significant changes in DUSD+ TZC and DU + TZC group( P 〉 0.05). Number of positive neuron in DUSD group was significantly lower than NSSD group( P 〈 0.05), but DUSD + TZC showed no difference( P 〉 0.05) Conclusions] SD can aggravate the impairment of the morphology and number of rat % somatostatin positive cortical neurons induced by DU particles instillation. TZC may protect rat's brain function by taking effect on somatostatin positive cortical neurons.
