涎腺肿瘤中增殖细胞核抗原,bcl-2,c-myc与细胞凋亡关系的研究

摘    要:目的:探讨涎腺肿瘤中细胞凋亡及其调控基因bcl-2,c-myc及增殖细胞核抗原(PCNA)间的关系,方法:采用脱氧核糖核酸末端转移酶介导的dUTP缺口末端标记(TUNEL)技术,免疫组织化学染色和HE染色对18例多形性腺瘤和52例恶性涎腺肿瘤中细胞凋亡和PCNA,bcl-2,c-myc蛋白表达进行观察和比较。结果:恶性涎腺肿瘤的凋亡细胞指数和增殖细胞指数明显高于多形性腺瘤(P<0.05),恶性涎腺肿瘤中bcl-2和c-myc蛋白表达率和表达强度明显高于多形性腺瘤(P<0.05),结论:细胞凋亡和调控基因异常在涎腺肿瘤中可能起重要作用,在涎腺肿瘤中,bcl-2和c-myc蛋白的共同作用可抑制细胞凋亡。

Abstract:Objective:In order to assess proliferating cell nuclear antigen (PCNA),bcl-2,c-myc and its relationship with apoptosis.we investigate PCNA,bcl-2,c-myc and its relationship with apoptosis in salivary gland tumor.Mothods:Apoptosis was detected in histological sections by the terminal deoxynucleotidyl transferase-mediated dUTP nick end labelling(TUNEL)technique,PCNA,bcl-2 and c-myc were detected by immunohistochemical staining in 18 cases of pleomorphic adenoma and 52 cases of malignant salivary gland tumor.Results:The apoptosis index and PCNA count were clearly higher in malignant salivary gland tumors than that in pleomorphic adenoma(P<0.05)?The positive rate and staining intensity of bcl-2 and c-myc oncoprotein in malignant salivary gland tumors were significantly higher than those observed in pleomorphic adenoma(P<0.05)?Conclusion:We conclude that the abnormal regulation of apoptosis may play an important role in salivary gland tumors;bcl-2 and c-myc oncoprotein may likely block apoptosis in salivary gland tumors?
