家蝇乙酰胆碱酯酶基因的克隆与序列分析

摘    要:目的克隆家蝇Ace基因并进行表达,并对家蝇AChE序列进行分析,为利用基因工程进行酶改造提供必要的参考. 方法结合已有的文献资料,利用生物信息学方法对家蝇AChE的序列进行分析,包括密码子偏爱性、系统发生、三维结构预测等. 结果成功克隆家蝇Ace基因,并利用同源模建方法获得了家蝇AChE的三维结构,以果蝇AChE为参考,判断了家蝇AChE应属于AChE 2家族. 结论根据所得家蝇AChE的三维模型从理论上分析了因AChE中一个氨基酸突变导致家蝇产生有机磷抗性的可能原因,并为半理性改造家蝇乙酰胆碱酯酶提供了三维模型.

Abstract:Objective To clone acetylcholinesterase(AChE) gene from Housefly (Musca domestica) in order to provide necessary reference for genetic engineering study. Methods The sequence of AchE was, by means of bioinformatics, analyzed including codon bias, systemic genesis, predicting of three-dimentional structure by using bioinformatics.Results AChE gene from housefly was cloned and AChE three-dimentional structure was obtained by homologous modeling. AChE,belonged to AChE 2 family, from fruitfly was taken as reference. Conclusion Based on housefly's AChE three-dimentional structure it is theoretically indicating the possibility that a mutation of an amino acid in Housefly's AChE can lead to the resistance of housefly to organic phosphorus pesticide.
