紫草素对肝癌细胞增殖的影响

摘    要:目的研究紫草素对肝癌细胞株HepG2增殖和表达增殖诱导配体(A proliferation-inducing ligand,APRIL)水平的影响,并与化疗药顺铂进行对比。方法MTT法检测紫草素对HepG2细胞增殖的抑制作用,采用免疫组化法和实时荧光定量PCR法证实肝癌细胞株HepG2上APRIL的表达,实时荧光定量PCR法检测加入不同终浓度的紫草素和顺铂后24h、48h和72h HepG2细胞表达APRIL mRNA的水平。结果紫草素对HepG2细胞有明显的增殖抑制作用,呈时间和剂量依赖性,HepG2细胞上有APRIL的表达,加入不同浓度的紫草素和顺铂后,HepG2细胞A-PRIL mRNA的水平均逐渐升高,至72h表达最高并与空白对照相比均有显著差异(P〈0.05)。结论紫草素可抑制肝癌细胞的增殖,但与顺铂一样,残存癌细胞有因APRIL表达升高而增殖能力增强的潜在趋势。

Effects of shikoni on the proliferation of hepatocellular carcinoma cells

Abstract:Objective To investiate the effect of shikoni in comparison with cisplatin (DDP) on the proliferation and expression of a proliferation - inducing ligand (APRIL) in hepatocellular carcinoma HepG2 cells. Methods MTT colorimetric assay was used to examine the growth inhibition of Shikoni in HepG2 cells. The protein and mRNA level of APRIL in HepG2 cells were determined by immunohistochemistry and real-time fluorescence quantitative PCR (RFQ-PCR). HepG2 ceils were treated with Shikoni and DDP at various concentrations for 24 h, 48 h and 72 h. The changes of APRIL mRNA level were analyzed by RFQ - PCR. Results Shikonin significantly inhibited the growth of HepG2 cells in a time and concentration - dependent manner. The expression of APRIL was strong in HepG2 cells. When treated with different concentration of Shikonin and DDP, the mRNA level of APRIL in HepG2 cells both raised gradually and reached the highest levels 72 h after treatment, which were significantly higher than that in blank control group (P 〈 0.05). ConeIusion Shikonin can inhibit the growth of hepatocellular carcinoma cells. On the contrary the proliferation ability of survival cells would rise because of the impact of APRIL after HepG2 cells were treated with Shikonin and DDP.
