摘    要:目的探讨硒代蛋氨酸和亚硒酸钠抗EV71病毒感染的效果,解析硒蛋白在机体抗感染的免疫作用。方法以非洲绿猴肾细胞(VERO细胞)和ICR小鼠为研究模型,用体外细胞培养和小鼠的动物实验来探讨不同含量的硒代蛋氨酸和亚硒酸钠对EV71在VERO细胞中和ICR小鼠体内增值的抑制作用,用荧光定量PCR方法定量分析病毒载量。结果 2μmol/L和3μmol/L的硒代蛋氨酸在5d内能有效抑制EV71在VERO细胞中的增值。而用5mg/L硒代蛋氨酸饲养的小鼠产下的幼鼠经EV71感染后,肌肉组织中的EV71核酸载量也比对照组显著降低。结论硒代蛋氨酸在体外和体内都能够有效抑制EV71复制,暗示硒代蛋氨酸可能参加了机体的免疫应答抗EV71的感染。

Effect of selenomethionine and sodium selenite on proliferation of EV71 in vitro and in vivo

Abstract:Objective To investigate the anti-infectious effect of selenomethionine(Se-Met) and sodium selenite on Enterovirus71 (EV71) and the immunization of selenoprotein. Methods Following pre-treated with Se-Met at different doses or sodium selenite, VERO cells and ICR mice were infected by EV71 to obtain the inhibition potential on proliferation of EV71 in vivo and in vitro. The content of EV71 was monitored by Real-Time PCR. Results The replication of EV71 was suppressed in VERO cells pretreated with 2μmol/L and 3μmol/LSe-Met in 5 days. Moreover, the content of EV71 was statistically decreased in the muscle of the offspring of ICR mice which were administered 5mg/L Se-Met. Conclusion Se-Met could suppress the replication of EV71 in vivo and in vitro, implying that Se-Met might play an important role in the immunization against EV71.
