三磷酸腺苷氯化镁对肺腺癌细胞的抗癌作用

摘    要:目的]探讨三磷酸腺苷氯化镁对肺腺癌细胞的抗癌作用。方法]人肺腺癌细胞系(A549)被分成4个组,每组每天在新换上的培养液中加入相应的药物,培养4d后用细胞记数方法观测其对癌细胞生长的抑制情况,再用扫描电子显微镜及透射电子显微镜观察用药后细胞表面形状及内部结构的变化。结果]在三磷酸腺苷氯化镁组和三磷酸腺苷组中癌细胞的生长受到显抑制,且有大量的细胞死亡,而加药后前2d三磷酸腺苷氯化镁的抑制,杀伤癌细胞作用较三磷酸腺苷明显,氯化镁组癌细胞的增殖未受到抑制,在电子显微镜下观察到,三磷酸腺苷组癌细胞的微绒毛变短,数目减少,核仁呈环状;三磷酸腺苷氯化镁组癌细胞的微绒毛变得极短,数目明显减少或消失,细胞表面光滑,核仁呈环状;氯化镁组无变化。结论]三磷酸腺苷氯化镁对肺腺癌细胞具有抑制及杀伤(细胞毒)作用,因为氯化镁与三磷酸腺苷结合形成的活性复合物增强了三磷酸腺苷的抗癌作用。

Abstract:OBJECTIVETo investigate the anticancer function of adenosine triphosphate magnesium chloride(ATP-MgCl 2) on lung adenocarcinoma cells.METHODSHuman lung adenocarcinoma cell (A549) was divided into four groups. Each group was added with corresponding drug every day to the newly changed culture medium. After four days culture, the inhibiting rate of cancer cells was measured by cell counting and the change of exterior and interior cellular structure was observed by electron microscope.RESULTSThe growth of cancer cells was inhibited markedly in ATP-MgCl 2 group and ATP group, lots of cells were ultimately killed. ATP-MgCl 2 showed higher growth inhibition rate and cytotoxic activity than ATP in the first two days. In MgCl 2 group, the growth of cancer cells was unaffected. With electron microscope, it was observed that in ATP group the microvilli of cancer cells were shortened and reduced remarkably, the nucleoli became annular. In the group of ATP-MgCl 2, the microvilli of cancer cells were shortened significantly and the number reduced significantly or even disappeared, and the surface of cells became velvet, and nucleoli became annular too. CONCLUSIONATP-MgCl 2 not only induces differentiation, but also shows a marked inhibitory and cytotoxic activity on human adenocarcinoma cell line (A549), because MgCl 2 can form an active compound with ATP, which increases the anticancer function of ATP.
