姬松茸多糖对哮喘模型小鼠气道炎症和Th2类细胞因子水平的影响

摘    要:目的]探讨姬松茸多糖对哮喘模型小鼠气道炎症和Th2类细胞因子水平的影响.方法]取雌性Balb/c小鼠40只,随机分为正常对照组、哮喘模型组和姬松茸多糖小、大剂量组,姬松茸多糖小、大剂量组分别灌胃给予姬松茸多糖200,400mg/kg.采用酶联免疫吸附法和蛋白质印迹法检测支气管肺泡灌洗液(BALF)中白细胞介素(IL)-4,IL-5,IL-13含量,并观察BALF中炎症细胞计数和肺组织病理学改变.结果]与正常对照组比较,哮喘模型组小鼠BALF中炎症细胞计数及IL-4,IL-5,IL-13水平均明显升高(P<0.05).与哮喘模型组比较,姬松茸多糖小、大剂量组BALF中炎症细胞计数及IL-4,IL-5,IL-13水平均明显降低(P<0.05).姬松茸多糖可明显减轻哮喘模型小鼠肺组织病理学改变.结论]姬松茸多糖具有抗哮喘作用,其机制可能与下调Th2炎症细胞因子、减少炎症细胞浸润有关.

Abstract:OBJECTIVE To study the effects of agaricus blazei polysaccharide(ABPS)on airway inflammation and Th2 cytokines in asthmatic mice model.METHODS40 female Balb/c mice were randomly divided into 4groups of control,asthma,low and high dose of ABPS,and the mice in low and high dose groups of ABPS were given by gavage with 200 and 400mg/kg of ABPS,respectively.The concentrations of IL-4,IL-5and IL-13 in bronchoalveolar lavage fluid(BALF)were measured by ELISA,and the number of inflammatory cells in BALF and histropathology changes were observed in lung tissues of mice.RESULTSThe number of inflammatory cells and the concentrations of IL-4,IL-5and IL-13 in BALF were significantly higher in asthma group than in control mice(P〈0.05),and obviously lower in low and high dose groups of ABPS than in asthma group(P〈0.05),and ABPS improved significantly the histopathological damage of lung tissues in the asthma group.CONCLUSIONABPS has the anti-asthma effect,and its mechanism may be relative to down regulation of Th2 cytokines and decrease of inflammatory cell infiltration.
