阿司匹林对乳腺癌细胞株的抑制作用

摘    要:目的 :观察阿司匹林体外抗癌作用并初步探讨其作用机理。方法 :将人乳腺癌细胞株 (MCF - 7)及小鼠乳腺癌细胞株 (HC - 11)与不同剂量阿司匹林共同培养 ,于培养 2 4h、48h、72h观察阿司匹林对两细胞株数目的影响 ,并以流式细胞术分析培养细胞不同时期细胞比例。结果 :阿司匹林对MCF - 7及HC - 11均有不同程度的生长抑制作用 ,且此作用与阿司匹林用量有明显量效关系 (72hIC50 分别为 10 1 6 2 μmol/L、5 3,2 0 μmol/L) ,共同培养 72h ,阿司匹林可引起MCF - 7细胞株G0 -G1期停滞 ,S期细胞明显减少 (11 8± 2 3) % ,(6 3±1 9) % ,P <0 0 5 ]。结论 :阿司匹林对人及小鼠乳腺癌细胞株有明确的抑制作用 ,其抑制作用是通过影响细胞周期进程 ,抑制细胞增殖 ,诱导分化而产生。本研究结果提示阿司匹林在乳腺癌的化学预防方面有相当的应用前景。

Abstract:Objective:The objective of the experiments reported here was to investigate the effect of aspirin on breast cancer cells and to study its mechanism.Methods: Human breast cancer cells (MCF-7) and mice breast cancer cells (HC-11) were co-incubated with aspirin in different doses (10 mmol/L,30 mmol/L,60 mmol/L,120 mmol/L).The numbers of cells were calculated after 24h,48h,72h treatment.Cytometry was employed to analysis the MCF-7 cells in different phases of cell cycle after 72h treatment.Results:A concentration-dependent reduction in cell number was observed in response to aspirin,which prevents MCF-7 cells from progressing from G0-G1 to S phase [(11.8±2.3)% vs.(6.3±1.9)%,P<0.05]. Conclusion: These data indicate that aspirin has an inhibitory effect on MCF-7 and HC-11,thus leading to halt in cell cycel progression, which may be responsible for the antineoplastic action of aspirin.
