燃煤型氟中毒对大鼠睾丸生精细胞凋亡的影响

摘    要:目的:观察燃煤型氟中毒对大鼠睾丸生精细胞凋亡的影响。方法:5月龄SD雄性大鼠18只,按体质量随机分为染氟组、染氟+大豆组和对照组,各组大鼠自由饮用自来水(含氟〈0.5 mg/L);各染氟组采用含氟量为69.33 mg/kg的煤烘玉米饲食,染氟+大豆组的大豆重量比为20%,对照组采用含氟为9.37 mg/kg的常食玉米饲食,实验期间动物自由进食、进水;染氟第90天,处死大鼠,使用H-E染色法和TUNEL方法对大鼠睾丸切片进行染色,观察细胞凋亡情况,结果:染氟组生精细胞凋亡个数高于对照组(278.17±25.47)个,(129±21.02)个,P〈0.05];染氟+大豆组生精细胞凋亡个数高于对照组(141.5±5.24)个,(129±21.02)个,P〈0.05];染氟+大豆组生精细胞凋亡数低于染氟组(141.5±5.24)个,(278.17±25.47)个],P〈0.05。结论:燃煤型氟中毒可造成大鼠生精细胞凋亡增加,增加营养后可有效减少氟中毒造成的生精细胞的凋亡。

Abstract:Objective: To observe the effect of coal-burning caused fluorosis on apoptosis of sperma-togenic cells in adult male rats.Methods: Eighteen healthy SD adult male rats aged 6 months were randomly divided into 3 groups(6 in each group): Control group(group C,fed with normal maize containing 9.37 mg/kg of fluorine),fluoride group(group F,fed with coal baked maize containing 69.33 mg/kg of fluoride),and fluoride plus soybean group(group S,fed with maize as group F plus 20% soybean).All the rats drank tap water in which sodium fluoride concentration was less than 0.5mg.Rats in 3 groups were killed in 90 days,and morphological changes of their testis were observed under light microscope after H-E staining.The number of apoptosis cells(NAC) in testis were detected by TUNEL assay.Results: NACs of group F(278.17±25.47) and group S(141.5±5.24) were bigger than that of group C(129±21.02)(P〈0.05),and NAC of group F was bigger than that of group S(P〈0.05).Conclusions: Coal-burning caused fluorosis can lead to high apoptosis of spermatogenic cells of rats,and adding nutrition can effectively reduce cellular apoptosis.
