天麻微卫星DNA分子标记与天麻素含量的关系

目的：探讨天麻遗传变异与其品质的关系，为优良种质的选育提供科学途径。方法：采用CTAB法提取天麻DNA，进行PCR扩增，用微卫星DNA分子技术（SSR）标记DNA；用高效液相色谱法检测不同种质天麻的天麻素含量，分析两者之间的关系。结果：出现A条带类型的样品天麻素含量较高，7EKYC、9EDF；24GBJ、22GZJ；1FDJ、2FLL；4VDF、6EKYW；18GWAC、8EWD，分别聚到一起，遗传距离较小。结论：特异引物扩增位点与天麻素含量有关，高天麻素含量与SSR高分子量条带有关，其关系有待进一步研究。

A Primary Study on the Relationship between Microsatellite DNA of Gastrodia elata Blume and Gastrodin Content

Objective： To investigate the associativity between the genetics and variation of Gastrodia elata Blume and its quality, so as to provide scientific path for good species selection. Methods： DNA of Gastrodia elata Blume was extracted by CTAB method, amplified by PCR and marked by SSR technique; Gastrodin content was detected by HPLC. The relation between SSR fingerprinting and Gastrodin contents was analyzed. Results： Gastrodin contents were relatively high in specimens with type A bands. Genetic distances between 7EKYC and 9EDF, 24GBJ and 22GZJ, 1FDJ and 2FLL, 4VDF and 6EKYW, 18GWAC and 8EWD were small. Conclusions： The sites for special primer amplification relate to Gastrodin content, and Gastrodin content relates to SSR fingerprinting, which needs further research.
