摘 要: | 目的 探讨多种细胞因子对高效抗反转录病毒治疗(HAART)的HIV/AIDS患者的免疫调节作用.方法 确诊HIV感染者或AIDS患者40例,CD4+T细胞数均<350/μl,接受HAART治疗,并分别于HAART治疗的0、12及24个月随访备血,健康对照组30例.ELISA检测血清和上清液细胞因子(IL-12、IL-21、IP-10和IFN-γ),流式细胞术检测CD4+T细胞、CD8+T细胞、CD4+CD25+FOXP3 Treg细胞.结果 未经抗病毒治疗的HIV/AIDS患者外周血中IL-12、IL-21和IFN-γ水平显著低于正常人,CD4+T细胞明显降低,HAART治疗后均不同程度升高;而IP-10、CD8+T细胞、CD4+CD25+FOXP3 Treg细胞显著高于正常人,HAART治疗后水平下降.CD4+T、CD8+T、CD4+CD25+FOXP3 Treg与各细胞因子水平有一定的相关性.结论 细胞因子可能和HIV/AIDS感染的致病机制相关,并可能参与了抗HIV病毒免疫重建和病毒控制过程.

Abstract: | Objective To investigate the regulation of some cytokines in human immunodeflciency virus (HIV)/acquired im- munodeficiency syndrome (AIDS) patients and their dynamic changes during 24 months of highly active antiretroviral therapy (HAART). Methods Forty Chinese HIV-1/AIDS adults, while their CD4^+ T cells were less than 350/ram^3 in peripheral blood, were accepted HAART. Vein blood was obtained from each patient at baseline (month 0), month 12 and 24 of HAART initiation and control groups. CD4^+, CD8^+, and CD4^+ CD25^+ FOXP3 Treg ceils were detected with flow cytometry. HIV RNA level was tested by real-time fluorescent quantitative polymerase chain reaction (RT-PCR). Meanwhile, enzyme linked immunosorbent assay (ELISA) was used to detect the serum levels of interleukin 12 (IL-12), IL-21, interferon-inducible protein 10 (IP-10), and interferon gamma (IFN-3,). Results IL-12, IL-21, IFN-γ, and CD4^4 T cells showed significantly lower, while the IP-10 and CD8^+ T, CD4^+ CD25^+ FOXP3 Treg cells significant higher in HIV infected patients than normal persons. After HAART initiation, IL-12, IL-21, and IFN-γ, levels were increased gradually, and were correlated positively with CD4^+ T cell counts; while, IP-10 levels and CD8^+ T, CD4^+ CD25^+ FOXP3 Treg cells were decreased gradually. CD4^+ T, CD8^+ T, and CD4^+ CD25^+ FOXP3 Treg cells were relative to some cyto- kines. Conclusions This study found a close relationships between cytokine levels and CD4 cell counts during 24-month HAART, and these cytokines might play appropriate roles in AIDS pathogenesis, and antiviral procedure and immune constitution. |
