载脂蛋白E基因剔除小鼠冠状动脉外膜炎症的演变过程

摘    要:目的:探讨载脂蛋白E基因剔除眼apolipoproteinEgene鄄knockout,apoE穴鄄/鄄雪演小鼠冠状动脉外膜炎性细胞在粥样硬化病灶发生、发展过程中的变化规律。方法:取30、60和120周apoE(鄄/鄄)小鼠心脏做连续切片,分别用HE染色、免疫组化、透射电镜鉴定冠状动脉外膜炎性细胞类型。结果:30周小鼠冠状动脉外膜以中性粒细胞浸润为主,构成比是66.04%,明显高于60周(6.49%)和120周(3.86%)的小鼠,P均<0.05;60和120周小鼠冠状动脉外膜以淋巴细胞浸润为主,构成比分别是59.74%和72.03%,明显高于30周(21.38%),P均<0.05。结论:在动脉粥样硬化病灶形成的过程中,外膜炎症经历了由急性炎症向慢性炎症病理变化的过程。

Abstract:Objective: To study the regulation and effect of the inflammatory cells at adventitia of coronary artery on the development and progress of atherosclerotic lesion in apoE(-/-) mice. Methods: Each heart of 30-, 60- and 120-week-old apoE(-/-) mice was cut consecutively and stained by HE staining, immunohistochemistry staining and electron transmission microscope to identify the types of the imflammatory cells infiltrated at adventitia of coronary artery. Results: There were a large number of neutrophils infiltrated at adventitia of coronary artery at age of 30 weeks with a constituent ratio of 66.04% which was significantly higher than those in 60- and 120-week-old mice (6.4% and 3.86%, respectirely, both P<0.05). The number of lymphocytes was increased in 60- and 120-week mice, with constituent ratios of 59.74% and 72.03% which were remarkably higher than that in 30-week mice(21.38%, both P<0.05). Conclusion: Adventitial inflammation of coronary artery undergoes a pathological process from acute to chronic stage in the formation of atherosclerotic lesions of apoE(-/-) mice.
