M21黑色素瘤细胞与淋巴管内皮细胞相互作用的形态研究

摘    要:目的:观察恶性黑色素瘤细胞转移时与淋巴管内皮的作用过程,探讨肿瘤淋巴转移的机理。方法:将M21黑色素瘤细胞接种在单层淋巴管内皮上,用相差显微镜和扫描电镜观察肿瘤细胞穿越淋巴管内皮的方式及两者的形态改变。结果:黑色素瘤细胞借胞膜突起与内皮粘附,粘附部位多位于内皮细胞连接处;粘附处的内皮收缩,肿瘤细胞穿越并在内皮下迁移。结论:肿瘤细胞的转移过程包括粘附、浸润和迁移;肿瘤细胞可以直接或通过内皮细胞间隙穿越淋巴管内皮向远处转移。

Abstract:Objective:To study the interaction between melanoma and lymphatic endothelial cell(LEC)and investigate the mechanism of tumor metastasis.Methods:M21melanoma cells were seeded on the confluent LECs monolayer and their morphological alternations were observed.Results:Most tumor cells adhered to LECs near the intercellular junction by pseudopodia.The adhesion made LECs retracted or de-stroyed,then tumor cells invaded the LECs monolayer and migrated under it.Conclusion:The steps of tumor metastasis include adhesion,invasion and migration,and tumor cells could metastasize through the lymphatic vessel by destroying the LECs or the intercellular gaps.
