摘    要:目的探讨乳酸-羟基乙酸共聚物(PLGA)导管修复猫动眼神经缺损时,局部应用睫状神经营养因子(CNTF)能否促进神经再生.方法家猫22只随机分为生理盐水(NS)组、CNTF组,对照组.右侧动眼神经制成4mm缺损,NS组、CNTF组分别用PLGA导管 NS、PLGA导管 CNTF修复神经缺损;对照组不修复.比较三组猫动眼神经再生情况.结果术后14周时,NS组有7/9只猫、CNTF组有8/9只猫的动眼神经功能有一定程度的恢复,大体标本、光镜及电镜显示动眼神经再生成功;CNTF组平均轴突直径较大、轴突通过率较高,与NS组比较差异有显著性(P<0.05).结论PLGA导管(明胶海绵、纤维蛋白胶)能有效修复猫颅内段动眼神经缺损;PLGA导管内局部应用CNTF能促进动眼神经再生.

Local administration of CNTF in PLGA conduit to repair oculomotor nerve defect in cat

Abstract:Objective To evaluate whether local administrate CNTF in PLGA conduit can enhance the oculomotor nerve regeneration in cat. Method] 22 cats were randomly divived into three groups, NS group, CNTF group and control group. In each cat, a 4mm nerve defect of the right oculomotor nerve was wade. The nerve defect in NS group were repaired with PLGA conduit, CNTF group were repaired with PLGA conduit and CNTF solution, nerve defect in control group were not repaired. After surgery, the pupillary light reflex and ocular movement were recorded every week. 14 weeks after surgery, the regenerating nerve were documented histologically and morphometrically in each cat. Result] 14 weeks after surgery, 7/9 cases of NS group, 8/9 cases of CNTF group showed partial functional recover of the oculomotor nerve. The anatomical continuity of the nerves in NS group and CNTF group were confirmed by histological examination.The ratios of nerve fiber transition and mean axonal diameter in CNTF group were superior to NS group (P <0.05). Conclusion] The PLGA conduit can sucessfully repair the oculomotor nerve defects of the cistern segment in cat. Local administration CNTF solution can enhance the nerve regeneration.
