摘    要:目的 应用酶消化法消化心肌组织、培养心肌细胞.方法 0.1%的胰蛋白酶重复消化乳鼠心肌组织多次,收集的细胞用含15%胎牛血清的MEM培养基混悬后过滤,纯化心肌细胞后置CO2培养箱孵育.结果 培养的心肌细胞4~6 h后开始贴壁生长,12~24 h明显增殖,3~4 d后细胞汇合成片.在倒置显微镜下心肌细胞呈圆形、梭形、多角形,伸出伪足,出现自发性节律性搏动.结论 应用酶消化法可简便、快速地获得大量活力高的心肌细胞.

Abstract:Objective To introduce how to culture cardiocytes after enzyme digestion of cardiac muscle. Methods Neonate rattus cardiac muscle was digested repeatedly by 0.1% trypsin. The cardiocytes were collected in MEM medium which contains 15% fetal bovine serum ,then suspended and filtered. The cardiocytes were placed into COz incubator to incubate after purification of them. Results Cardiocytes began to adhere and grow after 4-6 hours,to proliferate obviously after 12-24 hours, and to converge together after 3-4 days. The cardiocytes showed sphericalm, fusiform and polygon shape in the visual field of inverted microscope. All the cells stretched out parapodium and beated spontaneously and rhythmically. Conclusion A great quantity of vigorous cardiocytes can be cultured conviently and quickly by the application of enzyme digestion.
