苹果多糖对SW-1116结肠癌细胞凋亡的影响及机制研究

目的观察苹果多糖（apple polysaccharides,AP）对人结肠癌SW-1116细胞凋亡的影响及其机制。方法采用四甲基偶氮唑蓝（methyl thiazolyl tetrazolium,MTT）法观察AP对SW-1116细胞增殖的影响;以DNA ladder法和流式细胞手段研究AP对SW-1116细胞凋亡的作用;给予SW-1116细胞AP后,采用Western blot法检测凋亡相关蛋白caspase-3、caspase-8、caspase-9、Bax和Bcl-2的表达变化。结果 AP可明显抑制SW-1116细胞增殖,并诱导其凋亡;AP可促进SW-1116细胞caspase-3,caspase-8,caspase-9及Bax的表达,降低其Bc1-2的表达,且均表现为剂量依赖性。结论 AP可以通过线粒体和死亡受体途径诱导SW-1116细胞凋亡。

Effects of Apple Polysaccharides on the Apoptosis of Human Colon Cancer SW-1116 Cells and the Possible Mechanisms

Objective To investigate the effects of apple polysaccharides(AP) on the apoptosis of human colon cancer SW-1116 cells and the possible mechanisms.Methods The effects of different doses of AP on the proliferation of SW-1116 cells were observed by methyl thiazolyl tetrazolium(MTT) assay,and the effects of AP on the apoptosis of SW-1116 cells were investigated by DNA Ladder and flow cytometric analysis.After treated with AP,Western Blot was employed to determine the expressions of caspase-3,caspase-8,caspase-9,Bax and Bcl-2 in SW-1116 cells.Results AP could inhibit the proliferation of SW-1116 cells significantly in a dose dependent manner.AP could induce the apoptosis of SW-1116 cells by increasing the expression of caspase-3,caspase-8,caspase-9 and Bax,while decreasing the expression of Bcl-2 in SW-1116 cells.Conclusion AP can induce the apoptosis of SW-1116 cells through both the death receptor pathway and mitochondrion pathway.
