Abstract:Objective To compare the sensitivity and accuracy of fluorescent in situ hybridization(FISH) and chromogenic in situ hybridization (CISH) in detection of breast cancer HER2 gene. Methods HER2 gene amplification in 32 samples with breast cancer detected by CISH method was retrospectively tested by FISH method. Kappa test was employed to compare the agreement of the two methods. Results Ten samples with high copy amplification by CISH were all detected to be positive by FISH. Nine of 12 samples with low amplification by CISH were positive by FISH and the remaining 3 samples were negative. Nine of 10 samples with no amplification by CISH were detected to be negative by FISH and the remaining 1 sample was positive. The agreement rate of the two methods was 87.5%, K=0.724, P=0.000. Conclusion Both CISH and FISH technology can be applied to detect breast cancer HER2 gene. FISH method has higher sensitivity and accuracy.
