糖脉平对2型糖尿病大鼠血管中单核细胞趋化蛋白1表达的影响

摘    要:目的探讨糖脉平对2型糖尿病大鼠血管中单核细胞趋化蛋白1表达的影响。方法Wistar大鼠大剂量链脲佐菌素腹腔注射(STZ60mg/kg),造模前随机分为正常对照组、模型对照组、糖脉平组、二甲双胍组、玉泉丸组,成模后每2周称体重并剪尾测血糖,第6周处死取主动脉标本采用半定量RT-PCR法检测MCP-1mRNA蛋白表达。结果造模后,大鼠血糖明显升高,MCP-1mRNA的表达明显增高,经治疗后,糖脉平治疗组及二甲双胍组MCP-1mRNA的表达明显降低,与糖尿病模型组比较有显著差异(P〈0.01)。结论糖脉平能减少糖尿病大鼠MCP-1mRNA的表达,减轻血管内皮局部炎症反应,阻断和降低动脉硬化的形成。

Effects of Privet Tang Maipingfang on mRNA Expression of Blood Vessel Endothelium Monocyte Chemoattractant Protein-1 in Type 2 Diabetic Rats

Abstract:Objective To investigate the expression of blood vessel endothelium monocyte chemoattractant protein-1 (MCP-1) in type 2 diabetic rats following the treatment of Chinese medicine Tang maipingfang.Methods The type 2 diabetic Wistar rats were established by injecting high dose (60 mg/kg) of streptozotocin (STZ), and the successfully established rat models were randomly divided into model group (n=8), Privet Tang maipingfang group (n=11), Metformin group (n=11) and Yuquanwan group (n=10). Another 10 normal Wistar rats served as normal control. Their body weight and blood glucose were measured every two weeks. At the end of six weeks, all rats were sacrificed and their aortas were taken to detect the expression of MCP-1 by semi-quantitative RT-PCR.Results After successful establishment of rat model of type 2 diabetes, their blood glucose rose and their body weight decreased. After treatment, the expressions of MCP-1 in Privet Tang maipingfang group and Metformin group were significantly lower than that of model group (P〈0.01).Conclusion Privet Tang maipingfang can decrease the expression of MCP-1 mRNA levels, abate inflammatory reaction of blood vessel endothelium, restrain and decrease arteriosclerotic formation.
