摘    要:目的:通过骨折不愈合动物模型检测其外周血中PBMSCs的变化规律,深入研究骨折不愈合与PBMSCs的关系。方法:采集大鼠和人类的外周血进行PBMSCs和BMMSCs的细胞培养和用流式细胞仪检测CD34-/CD44+。结果:大鼠的单核细胞在人类BMMSCs在旋转生物反应器培养基在生物反应器中培养8d后,BMMSCs的细胞总数、CD34-CD44+和CD34+CD45+细胞分别较接种起始数量增加了9、29和8倍。结论:本文初步实验和已有研究证据证实在人、大鼠体内存在循环干细胞(PBMSCs),此类细胞可在体内或体外进行功能性成骨分化,测定循环间充质干细胞(PBMSCs)的量值变化来预测和早期诊断骨折不愈合的新方法和新标准。

The Preliminary Research and Explore About the Correlation between the Peripheral Blood Mesenchymal Stem Cells(PBMSCs)and the Animal Models of Nonunion

Abstract:Objective:To detect the variation o{ peripheral blood mesenchyrnal stem cells(PBMSCs)by animal models of nonunion, in-depth study the correlation between PBMSCs and nonunion. Methods: Collect rat and human peripheral blood to cell culture for PBMSCs and BMMSCs, then detect the CI~4^-/CD44^+ by flow cytometry. Results: Rat's monocytes and human BMMSCs cultured in rotating bioreactor after 8 days, the total number of BMMSCs cells, CD34^-/ CD44^+ cells and CI^4 +/CD45^+ceils increase in 9,29 and 8 times. Condusion: Preliminary experiments and research confirm that there has been PBMSCs in humans and rats. And these cells can be dfferentiation into osteocyte in vivo or in vitro. The nonunion can be forecast and diagnosed early by detection the change of the PBMSCs.
