摘    要:目的:研究小鼠骨髓基质干细胞的培养条件及其成为组织工程种子细胞的可能性.方法:取小鼠股骨骨髓进行骨髓基质干细胞的原代培养,通过相差显微镜、光镜及扫描电镜观察,了解骨髓基质干细胞的生长情况及其特点.结果:刚接种的干细胞散在分布,胞体透亮,1~2 d后开始贴壁,1周后开始形成成纤维细胞样集落,大约2周长成单层,碱性磷酸酶染色阳性,细胞可分泌胶原,可形成钙结节.结论:小鼠骨髓基质干细胞可在体外进行培养增殖并具有组织工程种子细胞的特点.

Abstract:Objective: To study the culture condition of marrow stromal stem cell (MSC) of mice and its possibility for becoming the seed cells of tissue engineering.Methods:Primary culture of MSC,which collected from femoral marrow of mice,were performed.The cells were observed with phase contrast microscope,light microscope,and scanning electromicroscope. Results:The MSC were diffusible in distribution. After 1 to 2 days,cell body was transparant. After 1 week,the cells began adherent. Colony-forming units,fibroblastic,was observed and single layer was formed in about 2 weeks and the active reaction of osteoblastic alkaline phosphatase marker was seen,collegen secreted,calcium node produced.Conclusion:The MSC of mice can be cultured and proliferated well in vivo,and it has features of the seed cell for bone tissue engineering.
