脂质过氧化和抗氧化作用对培养人内皮细胞的影响——冷冻复型的电镜观察

摘    要:首次应用冷冻复型方法对培养人血管内皮细胞的内部结构进行观察,表明内皮细胞的冷冻复型样品所显示的细胞器结构清晰,立体感强;对膜性系统的观察结果与扫描电镜和透射电镜下常规样品观察结果相符,而在显示细胞表面的小孔和核膜孔方面优于以前的电镜方法。并观察了氢过氧化枯烯和硒作用下内皮细胞膜性系统的改变,进一步探讨脂质过氧化与抗氧化作用对细胞的影响。

Abstract:The main feature of freezing etching is that it is able to show the internal structure of biomembrane. This study was aimed at making the freezingetching sample of cultured endothelial cells and observing the influence of lipidperoxidation induced by cumene hydroperoxide and antioxidation of seleniumon the structure of biomembrane. The results showed that the steric structureof the treated endothelial cells could be observed clearly under TEM. Especially,this method was superior to the ordinary observation under SEM and TEM inobserving the nuclear pole and cell membrane poles. The structural changes ofmitochondria and endoplasmic reticulum, plasma membrane blebbing, appearedby the treatmant of cumene hydroperoxide and the above changs was reducedby selenium added.
